Objective: To analyze the expression profile variation of long noncoing RNA(lncRNAs)in primary gouty arthritis(GA).In further,to explore the possible role of long noncoing RNA in GA patients via the correlation among the lncRNAs/cytokines and the clinic examination indexes.Method: 1).The peripheral blood mononuclear cells from 3GA patients and 3health controls(HC)were collected to detected for differently expressed lnc RNAs by microarray.The role of lnc RNA was predicted with GO and pathway analysis;2).30 acute gouty arthritis(AGA),24 non-acute gouty arthritis(NAGA)and 24 healthy controls(HC)were enrolled to verified the result of microarray by real-time quantitative polymerase chain reaction(RT-qPCR);3).RT-qPCR and Elisa were employed to examine the expression of critical nodes in PBMCs and plasma;4).100ug/ml MSU stimulate the blood of NAGA and HC,Real-time quantitative polymerase chain reaction(RT-q PCR)and Elisa were employed to examine the expression of lncRNA and the critical nodes in PBMCs and plasma;Results: 1).Compared with HC,there were 1815 lncRNAs more than 2 times variation,while 875 were up-regulated,940 were down-regulated.Bioinformatics analysis found that GO term enrichment included protein binding,regulation of transcription,metabolism,signal transdyction,et al.Pathway analysis found that cytokines and inflammatory response,glucocorticoid receptor network may involved.The expressionof AJ227913,AK001903 and ENSG00000239182 in AGA and NAGA groups were obviously higher than HC group(P<0.05),The expression of lnc-CCDC64B-1:8 in HC group was obviously higher than AGA and NAGA groups.2).The expression of IL-8 in PBMCs and plasma show significant difference among AGA,NAGA and HC groups,respectively.The expression levels of AJ227913 was correlated with IL-8 in plasma of AGA and NAGA groups.The transcription levels of IL-8 was correlated with IL-8 in plasma of AGA and NAGA groups.3).The expression of AJ227913 in NAGA which were stimulated by MSU was significant different compared with the control group,but this trend can not be found in HC group.The expression of IL-8 in plasma of the NAGA and HC which were stimulated by MSU was significant different,but this trend can not be found in mRNA in HC.The expression of IL-8 in mRNA of NAGA which were stimulated by MSU was significant different.4).The expression levels of AJ227913 was correlated with UREA(r=0.608,P<0.05)、CREA(r=0.337,P<0.05)、CYSC(r=0.422,P<0.05).Conclusion: 1).Several expression of lncRNAs was changed significantly in GA patients in comparision with HC.The level of AJ227913、AK001903、ENSG00000239182 was higher in GA than that in HC group;The expression of lnc-CCDC64B-1:8 in GA was significantly lower than that in HC group;2).Altered expression of AJ227913 might be involved in the inflammatory process of gout,and may also open another perspective to interpret the pathogenesis of gout.
