Study On Immuno-inflammatory Differences Between The Hepatitis B And C Genotype In The HEK-293T Cells

Aims:By comparing the immuno-inflammatory responses differences between HEK-293 Tcells which were infected by different gene segments of HBV,further clarifying the correlation between the genotype of HBV and disease course of hepatitis B.It is a complementary for HBV pathogenesis and offers direction for clinic treatments.Methods:Random selection 254 samples were HBV-DNA positive patients infected with chronic heptitis B virus from January 2015 to September 2015 visits in nanchang ninth people’s hospital,taking no account of ages 、genders and others.Excluding other illnesses and never receive antiviral treatment.Disease diagnosis in 2015《The diagnosis of chronic hepatitis b prevention》 guide standards.They were divided into 2 groups,with each group of 15 samples.15 cases were B genotype of hepatitis B virus,other were C genotype of HBV,which were identifyed by nested PCR with multiplex pairs of primers,are randomly selected.The s and c gene fragements were amplified through the HBV-DNA extracted from serum by PCR.The PCR products were recombined with T vector and pEGFP-C1 in order,then coexpressing in HEK-293 T cells,detecting the cytokine level,such as TFN-αand IL-1β.Results:IL-1βcytokine level in the cells coexpressed of s gene fragement from genotype B of HBV was143.89±46.48pg/ml,TNF-α cytokine level was 7.97±1.03 pg/ml.IL-1βcytokine level in the cells coexpressed of s gene fragement from genotype C of HBV was 40.96 ± 29.29 pg/ml,TNF-αcytokine level was 4.22 ± 0.56pg/ml;IL-1βcytokine level in the cells coexpressed of c gene fragement from genotype B of HBV was 253.62±65.12pg/ml,TNF-α cytokine level was 8.65±1.07pg/ml.IL-1βcytokine level in the cells coexpressed of c gene fragement from genotype C of HBV was35.78±19.31 pg/ml,TNF-α cytokine level was3.39±0.33pg/ml.Conclusions:The circular plasmids including s or c gene from genotype B of HBV can express higher cytokine level than genotype C of HBV.The pathogenesis of relation between genotypes of HBV and clinical manifestation of hepatitis B may be due to it in some sense.Genotype B and C of HBV may have some different response pathway.It led to the difference secretion levels between varies cytokines.The experiment helps us use another point to know the HBV.
