Effects Of Shexiang Baoxin Pill On Myocardial Apoptosis And Its Relationship With Bcl-2 Signaling Pathway In Rats With Heart Failure

Objective: To establish the rat model of heart failure,observe the effect of Shexiang Baoxin Pill on the function of heart of HF rats by pathological analysis and further to explore its mechanism.Methods: abdominal aorta constriction was used to establish rat model of heart failure.24 rats were weighed,numbered and randomly divided into 4 groups as follows: normal control group(NC),heart failure group(HF),Benner Pury group(BP)and Shexiang Baoxin Pill(SBP).After 8 weeks of continuous intragastric administration,the rats hearts were taken out to observe the appearance,and isolate left ventricular wall and weighed.The HE,MASSON and TUNEL staining were carried with paraffin section of ventricular wall.The protein expression levels of Bcl-2 and Bax were detected by Western blotting.Results:1.Compared with the NC group,heart mass index(p<0.05)and left ventricular mass index(p<0.05)in HF group were increased significantly,while in BP group and SBP group compared to HF group were decreased significantly(p<0.05).But the difference between with BP group and SBP group was not statistically significant(p>0.05).2.Pathological staining showed that,compared with the NC group,HF group showed obvious injury,myocardial fiber hypertrophy,collagen fiber hyperplasia,severe myocardial fibrosis,myocardial cell apoptosis(p<0.05);Benner Pury and SBP intervention group were able to different degrees of protection of myocardial tissue,inhibit myocardial fibrosis,reduce myocardial apoptosis(p<0.05 vs HF group).3.Compared with the NC group,the expression of Bcl-2 protein in HF group was significantly lower(p<0.05)while the Bax was upregulatedd(p<0.05).In BP or SBP groups,Bcl-2 were significantly increased(p<0.05),Bax expression was significantly reduced(p<0.05).The comparetion between group of BP and SBP showed no significant difference(p>0.05).Conclusion:1.Shexiang Baoxin pill can significantly improve the rat ventricular wall thickening,improve ventricular remodeling,reduce myocardial fiber damage,inhibit myocardial fibrosis,reduce the number of myocardial cell apoptosis,and play a positive role in the protection of heart failure.2.Bcl-2 signaling pathway may be involved in the inhibition of myocardial apoptosis and protect myocardial tissue of Shexiang Baoxin Pill,at least in relation to the up regulation of Bcl-2 and down regulation of Bax protein.
