In this study,the seeds of Psoralea corylifolia L.were chossen as a model drug to study the chang of chemical compositions before and after salt-roast processing,and characteristic fingerprint was constructed.First,according to the Chinese Pharmacopoeia,10 batches of different regions of psoralea were salt-roast processed.All the samples were prepared by using methanol extraction methods.Then a simple high-performance liquid chromatography method was established for analying of the main Chemical composition of psoralea,including psoralen,isopsoralen,isobavachin,neobavaisoflavone,bavachin,corylin,broussochalcone B,psoralidin,bavachinin,isobavachalcone,corylifol A,bavachalcone and bakuchiol.The change of the main ingredient of psoralea was compared before and after salt-roast processing.The content of total coumarins has no significant change,and the content of bakuchiol was reduced slightly.However,the content of total flavonoids content has been slightly rised.The chemical ingredients change of psoralea is not obvious before and after salt-roast processing.The correlation between compositions variation and functional variation of psoralea needed further study.Further,we studied the salt-roast processing impact on the efficacy of psoralea.The traditional Chinese medicine theory revealed that salt-roast could lead to drug treat on kidney meridian,enhance the effect of warming kidney and activating yang.Osteoblast model was selected for researching the correlation between the salt-roast processing and the osteoblast activity of psoralea.Its activity of stimulating osteoblasts proliferation and differentiation was measured by MTT and ALP activity assay.The result exhibited that serum containing scurfpea fruits had a significant osteoblasts proliferation and differentiation stimulating activities,and serum containing the salt-roast processing scurfpea fruits osteoblasts activity was exceed than the row samples.In addition,the osteoprotective effect of the extract of scurfpea fruits were investigated by using an ovariectomy induced osteoporosis model.The bone mineral density,biomechanical strength and bone microarchitecture was tested and surem biochemical in serum were determined.The results indicated scurfpea fruit had a good therapeutic effect for the treatment of osteoporosis rats induced by ovariectomized and after salt-roast processing the anti-osteoporosis efficacy of scurfpea fruit was significantly enhanced.All the results suggested that there was correlation between the salt-roast processing and the anti-osteoporosis efficacy of scurfpea fruit.It provided a scientific basis for clinical trials of pieces of salt-roast processing.We studied the pharmacokinetic of the main coumarins in rats plasma.A sensitive,specific and rapid ultra high performance liquid chromatography tandem mass spectrometry(UHPLC-MS/MS)method has been established to determine psoralen and isopsoralen in rats plasma.To compare two kinds of coumarins in vivo pharmacokinetics of orally administered scurfpea fruit raw products and salt products.The results showed that the Tmax of both psoralen and isopsoralen exhibited different degrees of extension,and Cmax was higher than the raw group,AUC0-t increased in varying degrees.The results illustrated that the salt-roast processing promoted the absorption of psoralen and isopsoralen,which increasing the bioavailability of these two coumarins compounds.Futher,A sensitive,specific and rapid ultra high performance liquid chromatography tandem mass spectrometry(UHPLC-MS/MS)method has been established to determine psoralen and isopsoralen in rats tissues,including heart,lung,spleen,liver,kidney,ovaries and testes.To compare distribution in rats tissues of two kinds of coumarins after orally administered scurfpea fruit raw products and salt products.Both psoralen and isopsoralen distributed in rats tissuea rapidly 15min after the administration.The results illustrated that the salt-roast processing promoted the distribution of psoralen and isopsoralen in rats tissues,especially,in heart,liver,ovaries and testes.The salt-roast processing had not affected on chemical ingredients change of psoralea,but its efficacy was changed.Salt-roast processing enhanced the anti-osteoporosis efficacy of scurfpea fruit.The reason is due to the change of the sbsorption of activity composition.
