Study Of The Mechanism Of Yifei Jiedu Formula And Ddp Synergetic Effect On Human Lung Adenocarcinoma A549 Line Cells

ObjectiveTo reveal the synergetic mechanism through study the effects of Yifei Jiedu formula and DDP to cell proliferation,cycle and apoptosis of the human lung adenocarcinoma A549 line cells.MethodsApply serum pharmacology method to prepare the serum containing Yifei jiedu formula.Apply MTT assay to detect the inhibitory effect of serum containing Yifei jiedu formula,DDP and the combination of two drugs on the human lung adenocarcinoma A549 line cells.Use flow cytometry to analyze cell cycle and measure the apoptosis rate of the human lung adenocarcinoma A549 cell which was treated by serum containing Yifei jiedu formula,DDP and the combination of two drugs.Use immunocytochemistry to detecte the expression of CyclinD1,Bax and Bcl-2 in the human lung adenocarcinoma A549 cell which was treated by serum containing Yifei jiedu formula,DDP and the combination of two drugs.ResultsIt showed that the rate of inhibition of Yifei Jiedu formula low dose,middle dose and high dose to the human lung adenocarcinoma A549 cell was 14.47%,25.32%and 33.12%;They were compared with control group,P<0.05,It showed that Yifei Jiedu formula inhibit proliferation of the human lung adenocarcinoma A549 line cells and showed a dose effect relationship.Index of cooperation was greater than 1.15 when Yifei Jiedu formula middle dose and high dose comnbined with DDP within the concentration range of 2mg/L-16mg/L.It showed Yifei Jiedu formula can enhance the inhibition of the human lung adenocarcinoma A549 cell,and it showed significant synergies and efficiency.The synergism was the strongest when Yifei Jiedu formula drug serum high dose combined with DDP within 8mg/L,Index of cooperation is 1.27,the highest.But the effect that Yifei Jiedu formula drug serum low dose combined DDP is simple adding together.It can lowered IC50 of DDP level when Yifei Jiedu formula drug serum low dose,middle dose and high dose combined with DDP.The combination of Yifei Jiedu formula drug serum and DDP can significantly arrest G0/G1 the human lung adenocarcinoma A549 cell cycle,meanwhile the expression of CyclinD1 was down.The combination of Yifei Jiedu formula drug serum and DDP can obviously induce the human lung adenocarcinoma A549 cell apoptosis,at the same time,the expression of Bax was increased and the expression of Bcl-2 was decreased.ConclusionYifei Jiedu formula can inhibit proliferation of the human lung adenocarcinoma A549 line cell.The combination of Yifei Jiedu formula and DDP can inhance the inhibition of the proliferation of the human lung adenocarcinoma A549 line cell,and it showed significant synergies and efficiency.The combination of Yifei Jiedu formula and DDP can arrest G0/G1 of the human lung adenocarcinoma A549 line cell,Its mechanism may be downregulation expression of CyclinDl.The combination of Yifei jiedu formula and DDP can induce apoptosis of the human lung adenocarcinoma A549 line cell,the mechanism maybe stimulate the expression of Bax,inhibit the expression of Bcl-2.
