BackgroundThe investigation found that the prevalence of coronary atherosclerotic heart disease increased year by year,the trend is more and more obvious.There is increasing evidence that inflammation is involved in the development and progression of coronary atherosclerosis.Nicotinamide adenine dinucleotide phosphate oxidase(NOX)is an intracellular protein with oxidation activity,cardiovascular physiological and pathological processes of the important regulatory factor.NOX expresses in phagocytic cells,platelets and almost all of the cardiovascular cells,and participates in the regulation of cardiovascular function,such as cell differentiation,proliferation,apoptosis,aging,and inflammation induced oxidative stress.NOX and atherosclerosis,hypertension,cardiac hypertrophy and vascular remodeling,the formation of collateral,the relationship between the pathological process of heart failure and the cardiovascular events closely.In endothelial cells Nox2 increased superoxide compounds,promots macrophages in early vascular lesions of the aggregation,Nox2 can increase the activity of platelets,promote platelet aggregation and platelet dependent thrombosis.ObjectiveTo study the diagnostic value of coronary heart disease,as well as the relationship between the degree of coronary atherosclerosis by detecting the Nox2 protein expression level of platelet.Doing something good in the diagnosis as well as the prevention of coronary artery atherosclerosis disease.MethodsAccording to the criteria for the selection,81 patients,who met the criteria of WHO and untreated CHD admission and underwent coronary artery interventional treatment were included,time is between August 2015 and February 2016.All of these patients were from the first hospital attach to Xinxiang Medical University.According to the coronary angiography results,all patients were divided into normal group(A group,n=22)and Coronary artery lesions group(B group,n=59),According to the results of coronary angiography,the coronary artery lesions group was divided into mild stenosis group(B1 group,n=13),moderate stenosis group(B2 group,n=18)and severe stenosis group(B3 group,n=28).Collect and write down all of the clinical feature of these patinets,such as age,strait relative related family hypertension history.Then we immediately detected the level of high density lipoprotein cholesterol(HDL-C)from blood of those patients.And we also immediately detect the complement3(C3)and Complement4(C4)from blood of those patients.Western blot method to detect the venous blood platelet Nox2 protein expression levels.The consent is informed and singed in all of these patients.ResultsAfter compared to the Control group,hypertension and the platelet protein level of Nox2 were much higher then Coronary atherosclerosis lesions group(P<0.05).There was much lower of HDL-C when it was compared with Coronary atherosclerosis lesions group(P<0.05).Platelet Nox2 protein expression level in coronary artery disease group was much higher than that in normal group(P<0.05).There is difference of age in these two group(P<0.05).Compared with the normal group,there were significantly higher level of platelet Nox2,C3,C4(P<0.05).The platelet Nox2 level of Coronary artery lesions group was in some way related to C3,C4.The expression level of Nox2 protein in normal group was higher than mild coronary artery disease group(P<0.05).In coronary artery disease group,the expression level of Nox2 protein was highest(P<0.01)in patients with severe coronary artery disease.Conclusions1.The expression level of Nox2 protein may have diagnostic value for coronary heart disease,and it is related to the severity of coronary atherosclerosis.2.Platelet Nox2 and C3,C4 may have synergistic effect,inflammatory factor Nox2 and humoral immune mechanism may be involved in the occurrence and development of coronary heart disease.
