Objectives The myocardial ischemia reperfusion injury was given in male Wistar rats before thoracic epidural block treatment,then collect rat electrocardiogram and heart rate,mean arterial pressure,measure the time between serum cardiac troponin I?cTnI?content in myocardial tissue,detection of autophagy related protein?Beclin-1?expression and TTC staining of myocardial infarct size and explore the thoracic epidural block on rat myocardial ischemia reperfusion injury?MIRI?rats.Methods 1 Research Group Thirty healthy adult male Wistar rats were randomly divided into : sham operation group?sham group?,myocardial ischemia-reperfusion?I/R?group and thoracic epidural block?TEB?group,with 10 rats in each group.The rats in the TEB group and?I/R?group were given 0.5% of Ropivacaine?0.125ml·kg-1?and the same amount of normal saline respectively in the epidural space before ligation,before it,give the normal saline?0.125 m L·kg-1?in the epidural space.The rats in the TEB group and I/R group were given a thread operation on the heart left anterior descending coronary artery and given a ligation,40 mins later,the ligature was loosed and then the rats were filled again for 120 min;TEB rat after the method of thoracic epidural block,according to the chest muscles in rats and the increase of the surface temperature,make the left anterior descending branch ligation in rats,then irrigate,After which,we can conclude The model was prepared successfully according to the general color of the heart and ECG.2 Electrocardiogram?ECG?and hemodynamic detection : Use multifunctional biological acquisition system in the body surface of rats to connect the body,continuous inspection record each group before opening his chest?T0?,Left coronary artery ligation ?T1?,ischemia 20 min?T2?,40 min?T3?,120 min reperfusion?T4?of heart rate?HR?,mean arterial pressure?MAP?.3 ELISA method to determine the content of serum troponin I?cTnI?:Each rat before the open chest?T0?,ischemia?T1?,ischemia 20 min?T2?,40 min?T3?and 120 min reperfusion?T4?in venous blood,using ELISA method to measure serum cTnI levels.4 TTC staining:120 min reperfusion executed in rats,take heart,with ice salt water is rinsed clean,hemorrhage,put heart into-20 ?refrigerator frozen 20 min,myocardial slices cut into 2 mm thick,with TTC stain,use Image-Pro Plus6.0 Image analysis software?American Media Cybernetics?calculation and myocardial ischemia of danger area of infarction area.5 Western blot method to detect myocardial tissue autophagy related protein?Beclin 1?expression level:120 min reperfusion after heart,put after liquid nitrogen refrigeration,the ischemia of danger myocardial tissue extraction of histones,detection of myocardial tissue autophagy related protein?Beclin 1?expression level.6 Statistical analysis of data:Use SPSS21.0 statistical software analysis,measurement data with mean????±s?)said,the single factor analysis of variance AVONA,comparing mean using t test(P < 0.05 for the difference was statistically significant.Results 1 Compared with T0 and the same time points of sham group,the HR and MAP at T2T4 in I/R group and TEB group significantly decreased?P<0.05?,compared with I/R group,the HR and MAP at T2T4 in TEB group significantly increased?P<0.05?,Three groups when compared with the T0 T1 HR,MAP,no statistically significant difference?P > 0.05?.2 Compared with T0,the serum c Tnl level at T2T4 increased significantly in I/R group and TEB group?P<0.01?,compared with the sham group,the serum c Tnl level at T2T4 in I/R group and TEB group significantly increased?P<0.01?,there was no significant difference in the serum c Tnl level between I/R group and group at T2T3?P>0.05?,while the serum c Tnl level at T4 in I/R group significantly increased compared with that in TEB group?P<0.01?,T0 when compare serum cTnI levels difference between three groups have no statistical significance?P > 0.05?.3 Compared with I/R group,the percentage of myocardial infarction area and myocardial ischemia danger area significantly decreased in TEB group?P<0.01?,and the rats in sham group had no myocardial infarction.4 Compared with the sham group,the Beclin-1expression in cardiac muscle was significantly higher in I/R group?P<0.01?,which in TEB group significantly decreased compared with that of I/R group?P<0.01?.Conclusion The TEB can canalleviate MIRI in rats,which may be related with cardiomyocyte autophagy factordown regulation.
