In this study,Gymnadenia conopsea was selected as the research material.The extraction technology of polysaccharide from Gymnadenia conopsea and the effect of polysaccharide from Gymnadenia conopsea on anti-aging,relieving fatigueand,hypoxia tolerance were studied in this study,which would provide some theoretical basis for development and utilization and further study of Gymnadenia conopsea.1)The processing of polysaccharide extraction from Gymnadenia conopsea were optimized by the single factor analysis and Box-Behnken Design.According to Box-Behnken response,the best extraction conditions were: the extraction times of 2,the extraction temperature of 97?,the extraction time of 160 min,and the ratio of material to liquid of 1:75(g/mL).Under the optimized condition,the polysaccharides content of the extracted solution was 12.19 %.2)The effects of polysaccharide from Gymnadenia conopsea on T-SOD,CAT,GSH-PX activity and MDA concentration in serum,liver tissue and brain tissue of mice was investigated by establishing D-galactose induced subacute aging model.The results showed that,compared with the model group,the high-dose group of polysaccharide from Gymnadenia conopsea can improve the activity of T-SOD,CAT and GSH-PX in serum,brain tissue and liver tissue of mice(P<0.01,P<0.05)and reduce MDA content(P < 0.01,P < 0.05)and improve the body weight of model group(P<0.01)and improve the liver index,spleen index and thymus index of model group(P<0.01,P<0.05).Compared with the model group,the middle-dose group of polysaccharide from Gymnadenia conopsea can improve the thymus index of model group mice(P < 0.01),compared with others experimental groups,there was not significant difference among the others indexes(P>0.05).Compared with the model group,the low-dose group of polysaccharide from Gymnadenia conopsea be not significant difference in the liver index,spleen index and thymus index(P > 0.05).There were not significant differences in renal index,lung index and brain index among the experimental groups(P>0.05).3)Through the mice loading swimming and no loading swimming andatmospheric pressure hypoxia and sodium nitrite acute poisoning experiments to explore whether the polysaccharide from Gymnadenia conopsea can relieve fatigue and anti-hypoxia.The results showed that,compared with the normal control group,the high-dose group of polysaccharide from Gymnadenia conopsea can significantly prolong the loading swimming time and decrease the area under the curve of blood lactate and the contents of hepatic glycogen in swimming mice(P<0.01),but be not significant difference in the middle-dose group and the low-dose group(P > 0.05).Compared with the normal control group,the high-dose group and the middle-dose group and the low-dose group of polysaccharide from Gymnadenia conopsea can reduce the levels of BUN in the blood and prolong the survival time of acute poisoning of nitrite in mice(P<0.01,P<0.05).To sum up,polysaccharide of Gymnadenia conopsea can be used to delay aging as well as relieve fatigue and anti-hypoxia.
