The Preparation And Of Modified Total Alkaloid Fraction From Nux Vomica And Its Application In Nux Vomica Powder

Nux vomiea Powder is reeorded in "Pharmaeopoeia of Chinese Nation ".It has the functions of dispelling rheumatism and freeing passages in your body.It can be used treat buttock-ache,Iumbago,ache all over and limb atrophy,ete.The toxieity of Nux vomiea is violent,and the amount of poisoning is near the amount of treatment.The study of subject is for changing chemical composition of Nux vomiea,and increasing efficiency and decreasing toxicity through compatibility of Nux Vomiea Power.Firstly,we studied the feasibility of baking method instead of stir-baked in sand to process seed of Strychnos nux-vomica L.Make the content determination of brucine,strychnine and brucine-N-oxide by HPLC.The content of total alkaloids in the samples was determined by the acid dye colorimetry.HPLC was used to cluster analysis.we aimed to find a safety and feasibility method to process seed of S.nux-vomica.the extraction of the total alkaloid fraction(TAF)from nux vomica was established.Most strychnine was removed from TAF and the resulted modified total alkaloid fraction(MTAF)was obtained.Then asystematic determination of chemical composition and correlation studies were conducted:the determination of alkaloid composition of TAF and MTAF and fingerprint research.In contrast to TAF,the ratio of brucine to strychnine was adjusted from 2.0:1 to 1:2.1 in MTAF.Strychnine was removed by optimization to a great extent,but brucine and other alkaloids of Strychnos nux-vomica L.were substantially retained with little loss.Acute toxicity test was carried out to determine toxicity of MTAF and TAF by oral route.The results showed that LD50 of MTAF and TAF were 15.10 mg/kg and 4.76 mg/kg,respectively.The toxicity was reduced more than three times.We also evaluate the quality of earthworm and the other ingredients of earthworm.we compared the analgesic、anti-inflammatory and immune effect of MP、MP-TAF and MP-MTAF.Hot plate test and acetic acid-induced writhing response were used to evaluate all the chemical-,thermal-analgesic activity in those;xylene-induced ear edema was used to evaluate anti-inflammatory activity;using delayed-type hypersensitivity test for the model to evaluate the immune activity.The pharmacodynamic evaluation results show that,MP-MTAF has better analgesic,anti-inflammatory,immune activity in contrast to MP andMP-TAF.Analgesic experiment also showed that the the effect of MP-MTAF will deline if the alcohol extract earthworm instead earthworm.Then the pharmacokinetic properties Brucine,MP,MP-TAF and MP-MTAF after oral administration in mice were compared.It was obvious that brucine in MP-MTAF was absorbed more completely than it in MP and Brucine at the same dosage of brucine.The AUC0-t of MP-MTAF group was 1.71 and 1.24 times higher than the Brucine and MP group,respectively.The pharmacokinetic results showed that the the:the oral absorption of MP has better than MP,MP-TAF.Ultimately from this research,we first a new Preparation that was MP-MTAF which the pharmacodynamicand oral absorption are superior to MP.
