Studies On The Chemical Constituents And Determination Of Mentha Haplocalycis Herba

Mentha haplocalycis herba is the dried aerial part of Labiatae family.It is cool in nature and acrid in taste.Mentha haplocalycis herba has the effects of dispelling wind-heat,refreshing head and releasing measles toxin.Thus,it has mainly been used in clinical treatment of wind-heat cold,beginning wind heat,red eyes,inflammation of the larynx,sore throat,mouth ulcer,toothache,hives,and rubella in clinic practices.Mentha haplocalycis herba as a commonly used traditional medicine,in the calendar edition of Chinese Pharmacopoia,quality control indexes for the content of the oil.In order to detect the therapeutic material basis of Mentha haplocalycis herba,control the quality of medicinal materials,and ensure the safety of medicine usage,this study analyzed its chemical composition and devised its quality control method.The contents include:1 Studies on the Chemical Constituents of Mentha haplocalycis herba.In this study,18 compounds were isolated and purified from the Mentha haplocalycis herba by a combination of chromatographic techniques including silica gel,octadecyl silica(ODS),Sephadex LH-20 column chromatography,middle and low pressure preparative chromatogram and pre-HPLC.And 17 compounds were identified on the basis of physicochemical properties and spectral data(1H-NMR,13C-NMR,HMBC,HSQC,MS,and HR-ESI-MS).Those compounds were identified as follows:menthalignin(1),gentisic acid 5-O-β-D-(6’-salicylyl)-glucopyranoside(2),syringaresinol(3),succinic acid(4),syringic acid(5),5-hydroxy-6,7,8,4’-tetramethoxyflavone(6),hesperetin7-O-β-D-glucopyranoside(7),agasta choside(8),didymin(9),linarin(10),acatin(11),tilianin(12),5-hydroxyl-6,7,8,3’,4’-pentamethoxyflavone(13),5,6,4’-trihydroxy-7,8-dimethoxy flavones(14),betulinic acid(15),oleanic acid(16),and ursolic acid(17).Among them compound 1 is a new lignin named mentha lignin;compound 6 is a new natural products;compound 7 is separated from the family of Labiatae for the first time;and compounds 2、3 and 15 are firstly reported in Mentha genus.2 Study on the measurement methods for the contents of Mentha haplocalycis herba.An HPLC method was established for simultaneous determination of four flavonoid(linarin,acatin,didymin and 5,6-dihydroxy-7,8,3’4’-pentamethoxyflavone)in Mentha haplocalycis herba.Simple and sensitive,this method was applied to determining the contents of 10 batches of Mentha haplocalycis herba.An HPLC method was established for simultaneous determination of four phenolic acids(syringic acid,syringaresinol,caffeoyl glycolic acid and succinic acid)in Mentha haplocalycis herba.Simple and sensitive,this method was applied to determining the contents of 10 batches of Mentha haplocalycis herba.3 Studies on the HPLC Fingerprint of Mentha haplocalycis herba.This research establish a preliminary HPLC fingerprint of Mentha haplocalycis herba.,and employed the "TCM Fingerprint Similarity Evaluation Software(version 2004)," which is developed by Chinese Pharmacopoeia Commission,in evaluating the similarity of the 10 batches of Mentha haplocalycis herba.It is found that a total of 11 characteristic common peaks were identified,with the similarities ranging from 0.816 to 0.997.
