| Diabetes mellitus is a major chronic disease affecting a large population across the world.Glycyrrhizae Radix is a traditional Chinese herbal medicine distributed widely in the northwest part of China.The licorice roots have been used as flavoring and sweating agents,as well as demulcents and expectorants in the world for thousands of years without evidence of adverse effects or toxicity.It is reported that the EtOH extract of licorice had anti-diabetic action.Previously,licorice flavonoids is found possessing strong a-glucosidase inhibitory effect,and thus the licorice flavonoids dispersible tablets(LFT)were prepared and its anti-diabetic effect in Alloxan-induced diabetic rat and high fat diet-induced type 2 diabetes mouse were studied.Results showed that LFT had high anti-diabetic activity.The present study investigated the effect of LFT on serum glycose,blood HbA1C%,serum insulin and liver glycogen levels in STZ-induced diabetec mice,and we also developed a quality control standard of LFT.Results:(1)Study on the anti-diabetic effect of LFT in STZ-induced diabetic mice.After 21 days of treatment with LFT,serum glucose and HbA1C%levels of the LFT treated group were significantly lower than that of diabetic control group(p<0.001),whereas serum insulin and liver glycogen levels of the LFT group showed a increasing trend,but there was no significant difference between the LFT and diabetic control groups.(2)Study on the quality control standard of LFT.We identified LFT through TLC,results showed that the method was specific without interference.The method of dissolution test for LFT was established:dissolving medium was distilled water;temperature:37 ℃;Paddle rate:100 r·min-1.At the same time,we developed both UV and HPLC determination methods to detect the content of effective constituent in LFT.Results showed that both of the methods were sensitive,reproducible and can be used for controlling the quantitative determination of LFT. |
