Gene therapy is guide the DNA into the target cells to cure the disease,so that the exogenous genes to correct or repair defects, shut down and inhibit abnormal expression of genes. Achieving the goal of cure the disease. As a new method for the treatment of cancer,this way has made significant achievements in chinical areas. Poly(V,N-dimethylaminoethyl methacrylate) (PDMAEMA) is a good modal for research the relationship between the cell transfection efficiency and the polymer structure. More attention have paid in the genetic carrier field. And with the excellent genetically modified performance, it is expected to become a breakthrough in the field of gene transfer in the future.In this paper, a series of PDMAEMA homopolymer and PEGylated copolymer with different degree of polymerization were synthesized by atom transfer radical polymerization (ATRP) in aqueous media firstly. By FT-IR spectrum and NMR spectroscopy tests, it confirm that homopolymer and copolymer with the structure of polymer and the relative molecular weight in line with the agreement. By Uv-Vis test, it shows that the homopolymer and copolymer all have a temperature dependent performance.And, I have studied a variety of factors effect the LCST. So, we can know whether the polymer LCST corresponding to the performance on this polymer as a genetic carrier.The program choosing human colon cancer cells (HCT-116) as the research object.Extracting the encoding protein Flag labels pCMV-3×Flag-LTA4H coli plasmid. Using the theory of unlike charge electrostatic, there are many nanoparticals by self-assembly. My samples were used in gene transfer, and compare to the Lipofectamine 2000 reagent (Life).And make a conclusion by cell transfection efficiency experiment and MTT test.The results shows that the PDMAEMA homopolymer with high transfection efficiency in vitro, and a certain cell toxicity. PEGylated can improve the application of gene carrier,although reduce transfection efficiency after introduce the PEG. At the same time,PEGylated can reduce cytotoxicity. More importantly, PEG can serve as a linker for the chain growth and provide a broad space for the vehicle development in the future.
