The Efficiency Evalution Of Jianpi Xiaoke Recipe For Newly Diagnosed T2DM Islet Function And The Effect Of The Expression Of DNase I In T2DM Rat Pancreas Islet

Ojective:To evaluate the clinical effect of Jianpi Xiaoke Recipe on newly diagnosed patients with type 2 diabetes islet functional improvement,and its implicat-ions for expression of DNase I in pancreatic tissue of type 2 diabetic ratsMethods:Clinical research:There are 70 patients with newly diagnosed T2 DM who have spleen deficiency,and retention of damp and hot.Randomly divided into treatment group(Jianpi Xiaoke Recipe and Glimepiride tablets)and control(Glimepiride tablets).To evaluate two groups of patients before and after treat-ment of blood glucose,blood lipid,glycosylated hemoglobin,fasting insulin,insulin resistance index,beta cell function index,body mass index and other indexes;Experimental research:Injected intraperitoneally with streptozoto-cin(STZ)Wister rats,in order to establish T2 DM rats model.To evaluate the effect of Jianpi Xiaoke Recipe on T2 DM rats weight,blood glucose and blood lipid,fasting insulin,insulin resistance index and islet beta cell function index,Morphological changes of pancreatic islets in rats by HE staining,Immunohistochemistrydetect the expression of DNase I in rat pancreas.Results:Jianpi Xiaoke Recipe can significantly improve the blood glucose,blood lipid,body mass index,increased insulin sensitivity,islet function with the newly diagnosed of T2 DM.Additionally,it can inhibit DNase I expression in pancreatic islets of T2 DM rats,to protect the islet cells.Conclusion: Jianpi Xiaoke Recipe can improve islet function with the newly diagnosed T2 DM patients and has higher safety.The mechanism may be related to inhibiting the expression of DNase I in pancreatic islets,which may play an important role in the protection of islet beta cells.
