Objective:To uncover the effect of asarum effective components(Methyleugenol)upon the animal epileptic behaviors.To prove methyleugenol have antiepileptic effect.Methods:(1)To observe animal epileptic behaviors on PTZ chronic epileptic model.Adult male SD rats were randomly divided into three groups:model group,methyleugenol group,VPA positive control group.Saline,methyleugenol(120mg/kg),sodium valproate(400mg/kg)were injected into the abdominal cavity everyday.One hour later,animals reseived a subthreshold-dose PTZ(40mg/kg,every day)to establish PTZ-kindlde epileptic rats model by intaperitoneal injection.To record the degree of epileptic seizures until the end of the 28 days.(2)To establish pilocarpine-Induced epileptic rat model.Adult male SD rats were injected intraperitoneally with pilocarpine(50mg/kg),then SD rats were randomly divided into three group:medol group,methyleugenol group,VPA positive control group which has appeared SRS.On the fourth day,saline,methyleugenol,sodium valproate were injected into abdominal cavity after model establishment.Then,continuous injection of 7days.Observe and record the degree of epileptic seizures and the SRS times.(3)Adult male rats were randomly divided with the same treatment,but the rats were injected for 7 days.The eighth day,animals were injected intraperitioneally with PTZ(60mg/kg),recording the latency to the first seizure and the field potential at the sametime.(4)Made the brain slices from the baby rats.Aplicated the patch clamp technology,to record the discharge frequency(AP)of hippocampal neurons after perfused by methyleugenol.Result:(1)In thePTZ-kindled epileptic rat modle,we observed that the kindling rate and the degree of attack were significantly lower,the latency was prolonged in methyleugenol group and VPA positive control group than that in medol group(P<0.05).But the methyleugenol group with the VPA positive group have no difference(P>0.05).Therefore,we infer that the methyleugenol has antiepileptic effect.(2)In the pilocarpine-induced epileptic model,we oberved that the number of SRS was decreased in methyleugenol group and VPA positive group.(3)Methyleugenol could inhibit the seizure and prolonged the latency of intensive discharge(P<0.05).But positive group than methyleugenol group and medol group have no difference(P>0.05).(4)Discharge frequency was significantly lower than before perfused by methyleugenol(P<0.05).Couclusion:Methyleugenol can inhibit epileptic seizures on PTZ chronic epileptic model and pilo-induced epileptic model.Methyleugenol can inhibit the latency of intensive dis-charge.In magnesium free artificial cerebrospinal fluid,discharge frequency of hippocampal neurons were reduced by perfused methyleugenol.Then,we infer that methyleugenol could antiepilepsy by acting on central nervous system.
