| Objective Detecting activity of patient with type 2 diabetic peripheral neuropathy(DPN)of peripheral blood mononuclear cells telomere length and telomerase reverse transcriptase(TERT),patients were antioxidants before and after treatment,we tried to look after pathogenesis of DPN in molecular level.Methods We choose select 80 cases aged 50-70 years adn diagnosed type 2 diabetes with and without peripheral neuropathy who hospitalized in the Department of endocrinology of Bao Tou Medical College first affiliated hospital from 2013 August to 2014 October and 40 cases of healthy persons as control group(N).We take inclusion criteria from 2013 China diabetes prevention guidelines in peripheral neuropathy(DPN),and all the selected diabetic patients were improved the nerve electrophysiological examination.Group DPN aged 50-70 for DPN-I,60-70 years for DPN-II.Application of real-time fluorescent quantitative polymerase chai detecting all samples of peripheral blood mononuclear cells of telomere length and telomerase reverse transcriptase(TERT)expression level,Changes of telomere length and TERT before and after treatment of antioxidant in DPN group,the DPN group,NDPN group between TEL,TERT and duration of diabetes,NCV,HbA1 C were compared.Evaluation of TEL,TERT could as a peripheral neuropathy in early diagnosis and evaluation of drug efficacy of clinical index Results using statistical software SPSS19.0 statistical data,the sample mean t test and single factor variance analysis method.Results 1,telomere length: N group >NDPN group >DPN group,DPN-I > DPN-II(P<0.05);TERT :group N < group NDPN < group DPN: DPN-I < DPN-II(P<0.05).2,By analyzing the correlation analysis of peripheral nerve conduction velocity in diabetic patients、TEL、TERT,We concluded that there was a significant positive correlation between each group NCV and TEL(0 < r <1,P<0.05),a significant negative correlation between each group NCV and TERT(-1< r <0,P<0.05).3,analyzing the correlation analysis of course、HbA1C and TEL、TERT,We concluded that there was not a significant correlation between that.4,The telomere length increased slightly after antioxidant treatment,but there is no statistical significance(P>0.05).Conclusion 1,Activity expression of telomere length and telomerase reverse transcriptase in peripheral blood mononuclear cells with,you can assess the damage neuropath.2,Type 2 diabetic peripheral neuropathy can lead to aggravation,telomere length shorter and telomerase reverse transcriptase activity increased.Statistical analysis was performed NCV and telomere length was positively correlated,NCV and telomerase reverse transcriptase activity was negatively correlated,Comprehensive analysis of the results,two activity was detected as the onset of diabetic peripheral neuropathy in type 2 and severity indicators.3,Affected by many factors,no significant correlation between the Course of disease and telomere length and telomerase reverse transcriptase activity.4,Natural telomere length in peripheral blood mononuclear cells expressed reduce and telomerase reverse transcriptase activity increased,this difference is statistically significant,age is one of the biological mechanisms explain the pathogenesis of DPN.5,After antioxidant therapy increased telomere length and decreased TERT,but the difference was not statistically significant,Shorter observation time there is a certain relationship... |
