| OBJECTIVE:To investigate the effect of deregulated expression of microRNA-187 on proliferation,invasion and migration of gastric cancer.METHODS:A total of 24 pairs of human gastric cancer tumor and matched non-tumortissues were collected and miR-187 expression was detectedand their relevance was analyzed.The expression of miR-187 and CD276 in human gastric cancer cell SGC-7901 were inhibited by transfection,and then the effect of SGC-7901 cell proliferation and invision-migratory was respectively detected by CCK-8 and Transwell assay.The expression of matrix metalloproteinase-9(MMP-9)was detected by Western blot.RESULTS:1.Real-time PCR showed that miR-187 expression in gastric cancer was increased,and low expression in paracancerous tissue(P<0.001).Western blot analysis confirmed that CD276 expression in gastric cancer decreased,and in the adjacent tissue high expression.The expression intensity of miR-187 was correlated with lymph node metastasis(P=0.046),TNM stage(P=0.015)and CerbB-2 expression(P=0.013)in gastric cancer,but not with age,sex,location of tumor,tumor size,depth of invasion and expression of Ki67(all P>0.05).MiR-187 expression in gastric cancer with lymph node metastasis more than 3,stage III and IV and CerbB-2 positive gastric cancer was significantly stronger than lymph node metastasis less than 3,stage I and II and CerbB-2 negative gastric cancer.The expression intensity of CD276 was correlated with lymph node metastasis(P=0.044),depth of invasion(P=0.042),TNM stage(P=0.001)and Cerb B-2 expression(P<0.001),but not with the patient’s age,sex,tumor location,tumor size and Ki67 expression(all P>0.05).CD276 was significantly lower than lymph node metastasis in tumor tissues with lymph node metastasis of T3,T3~T4,III and IV and CerbB-2 positive gastric cancer.T1~T2,I and II and CerbB-2 negative gastric cancer tissue.2.Compared with normal tissues,mi R-187 expression in stage I,II,III and IV gastric cancer tissue showed a progressive increase,respectively,the normal tissue of the stomach 20.3 times,3.5 times,16.3 times and 11.8 times(P<0.001);CD276 in stage I,II,III and IV gastric cancer tissue expression decreased,respectively,the normal tissue of the stomach 0.68 times,0.71 times,0.67 times and 0.61 times(P<0.01);The expression of miR-187 in gastric adenocarcinoma SGC was 5.5 times higher than that in gastric mucosa epithelial cells(P<0.001).3.The correlation coefficient R2 of miR-187 expression and CD276 expression in gastric carcinoma was 0.840,the expression of miR-187 in gastric cancer specimens could explain the variability of CD276 expression in 76.63%,another 23.37% variability can not be explained by miR-187 expression.4.Compared with the control group(miR-187 inhibitor-control group),miR-187 inhibitor(miR-187 inhibitor)SGC proliferative activity was(60.67±11.02)% lower than that of the control group(P = 0.003),and the invasive rate was(60.67±11.02)% lower than that of the control group P = 0.001)and the migration ability was(66.33±10.6)% lower than that of the control group(P <0.001);The expression of CD276 in miR-187 inhibitor group was increased by(1.047±0.06506)times(P <0.001)and the expression of MMP-9 was decreased by(0.2667±0.06028)times(P <0.001).5.Compared with the control group(CD276-siRNA-control group),the proliferative activity of CD276-siRNA(CD276-siRNA)was(1.823±0.2577)times(P = 0.002)in the control group and(1.843±0.3296)times(P <0.001)And the migration was 1.67 times(1.66±0.1453);The expression of CD276 in CD276-siRNA group was decreased by(54.67±4.509)%(P = 0.025)and MMP-9 expression increased by(0.4767±0.07234)fold(P = 0.038).CONCLUSION:1.MiR-187 expression in gastric cancer tissue and gastric cancer cell SGC was significantly higher than that in adjacent normal tissues and gastric epithelial cells,CD276 expression in gastric cancer and gastric cancer cell SGC was significantly lower than that in adjacent normal tissues and gastric epithelial cells.2.The expression intensity of MiR-187 and CD276 was correlated with lymph node metastasis,TNM stage,depth of invasion and CerbB-2 expression in gastric cancer.3.There was a linear correlation between gastric cancer tissue and adjacent normal tissue,MiR-187 and CD276,R2 = 0.84.4.Inhibiting the expression of miR-187 in SGC cells inhibited the proliferation of gastric cancer cells,and decreased the ability of invasion and migration;The expression of CD276 and the expression of MMP-9 were decreased in gastric cancer cells.5.Interleaving the expression of CD276 in SGC cells promotes the proliferation of SGC cells,Gastric cancer cells increased the ability of invasion and migration,MMP-9 expression increased. |
