Objective: To study the function of miRNA-9 on osteosarcoma MG-63 cells proliferation and apoptosis and the correlation with CXCR4 by improving the expression of mi RNA-9,to provide theoretical basis for the gene therapy of osteosarcoma.Methods: Osteosarcoma MG-63 cells are randomly divided into three groups:mi RNA-9group(transfected with miRNA-9 Oligo),NC group(transfected with miRNA negative contract)and blank group(transfected nothing).Observed the cell transfection effect by fluorescence microscope,Detected the expression of miRNA-9 and CXCR4 by real time PCR.Detected the CCK value(OD)at 24 h,48h,72 h and 96 h,then observed the cell growth curve.The apoptosis was measured by flow cytometry.Results:(1)After transfection of miRNA-9 Oligo,we observed the green fluorescence by fluorescence microscope in 70% osteosarcoma cells.(2)Compared with negative control group and blank group,the miRNA transfection group was higher expressed on mi RNA-9(P<0.01).(3)Osteosarcoma cells showed lower cell proliferation efficiency and higher cell apoptosis rotio on miRNA-9 higher expression groups(P<0.01).(4)The expression of CXCR4 mRNA was lower in miRNA-9 group compare with NC group and blank group(P<0.01).Conclusion:(1)High expression of miRNA-9 in osteosarcoma MG-63 cells can inhibit cell proliferation,promote cell apoptosis;(2)CXCR4 expressed in osteosarcomaMG-63 cells,the expression level of CXCR4 mRNA and mi RNA-9 is negative correlation in osteosarcoma MG-63 cells,the higher expression of miRNA-9 is lower expression of CXCR4 mRNA in MG-63 cells.CXCR4 gene probably involved in the miRNA-9 inhition proliferationof osteosarcoma MG-63 cell,and promote cell apoptosis.
