Radiation can induces oxidative stress and acute inflammation which may continue for a long time and lead to chronic inflammation.With increasing of cancer frequency,patients with cancer receive multimodality therapy to include surgery,radiation,and/or chemotherapy.Approximately 60% of all cancer patients undergo radiation as a component of their treatment at some time during the course of their disease.Ionizing therapeutic radiation is a potent physical entity with dramatic effects in both malignant and surrounding normal tissues.Untoward reactions to radiation therapy in normal tissues are most frequently classified as either acute reactions or delayed reactions.The mechanism of acute radiation injury is that the body is exposed to radiation,acting on the DNA molecule,which leads to the wrong combination of base pairs and molecular fracture;At the same time,it acts on the ionization of water molecules to generate reactive oxygen species,and the reactive oxygen species is active,which can react with biological macromolecules to cause radiation damage;Radiation can also change the activity of some enzymes,resulting in cell metabolism disorder,thereby changing the body’s immune,nervous and endocrine system.In order to find a radioprotectant of real worth,we should consider the following features,such as bioavailability,safety and toxicity,distribution,and free radical scavenging ability.Grape seed proanthocyanidins can remain in the plasma and tissues for up to 7–10 days and exert antioxidant properties with no toxicity.Compound raspberry seed powder contains a lot of proanthocyanidins,vitamin E,vitamin C and other antioxidants.Fifty wistar rats were randomized divided into five groups: normal control group,radiation group,vitamin group,grape seed proanthocyanidins group(GSPS group),compound raspberry seed powder group.Before radiation,vitamin E(30 mg/Kg)and C(50 mg/Kg)was given to rats in vitamin group,grape seed proanthocyanidins(125mg/Kg)was given to rats in GSPS group,compound raspberry seed powder(2.5 g/Kg)to those in compound raspberry seed powder group.The intragastric administration lasted for 7 days.On 8th day,a radiation of 8 Gy was offered to rats except control group.All rats were sacrificed 24 hours later,to detected WBC,PLT,superoxide dismutases(SOD),malonaldehyde(MDA),glutathione peroxidase(GSH-PX)and the damage of DNA using single cell gel electrophoresis technique.To investigate the damnification of the rat which is exposed to the X ray and the protective effect of proanthocyanidins and compound raspberry seed powder on acute irradiated rat.The results are as follows:1.Compared with the blank control group,irradiation group has the significant difference in the quantity of the WBC,RBC and PLT,the difference was statistically2.significant(t =7.842,3.985,P <0.01);Compared with irradiation group,vitamin group,compound raspberry seed powder group and grape seed proanthocyanidins group have the significant difference in the quantity of the WBC,RBC,and PLT,the difference was statistically significant(P <0.05).3.Compared with the blank control group,irradiation group SOD activity and GSH-PX activity decreased,MDA content increased,the difference was statistically significant(t =4.178,2.251,2.937,P <0.05);Compared with irradiation group,vitamin group,compound raspberry seed powder group and grape seed proanthocyanidins group SOD activity and GSH-PX activity were increased,MDA the content decreased,the difference was statistically significant(P <0.05).4.Compared with the control group,irradiation group DNA lymphocyte injury is more serious,there was statistical significance(t =3.997,P <0.05);Compared with irradiation group,vitamin group,compound raspberry seed powder group and grape seed proanthocyanidins group lymphocyte DNA damage was lower,the difference was statistically significant(P <0.05).5.Compared with the control group,irradiation group lymphocyte DNA serious damage,the comet head dark tail extension,vitamin group,compound raspberry seed powder group and grape seed proanthocyanidins group lymphocyte DNA damage lighter.Stated thus,raspberry seed powder or grape seed proanthocyanidins is effective in radiation protection.
