Objectives Early spontaneous abortion of embryonic villi(Early abortion villi)were detected by conventional chromosome karyotype analysis method and the whole chromosome aberration of DNA sequencing(NGS)technology,To explore the etiology of early spontaneous abortion genetics and the application of whole chromosomal aberration DNA sequencing were used to detect embryonic villi in early spontaneous abortion value.Methods 117 patients with early abortion clinic were collected from October 2015 to January 2017 in tangshan city maternal and child health care of obstetrics and gynecology and reproductive genetic families,NGS technique and chromosome karyotype were detected in the early spontaneous abortion villi collected by the woman's palace,and the two methods were used to detect the peripheral blood of 117 couples.The percentage of aberrant abortion NGS chromosome aberration was analyzed,and the distribution characteristics of abortion age in different pregnant women were analyzed.Results 1 In this study,NGS technique and chromosome karyotype analysis were detected in 117 cases of early abortion villi,and the abnormality rate of chromosome was higher than that of karyotype analysis(62.4%> 47%).The difference was statistically significant(P <0.05);The detection rate of chromosome aberration(2.6%)was less than that of karyotype analysis(4.3%),and the difference was not statistically significant(P> 0.05).2The NGS chromosome abnormality rate of ?35 years old group was higher than that of<35 years old group(74.1%> 52.4%),the difference between the two groups was statistically significant(P <0.05);The abnormal rate of NGS copy number variation was slightly higher than that of <35 years old group(31.48%> 25.39%),but there was no significant difference between the two groups(P> 0.05).The abnormality rate of NGS chromosomes in sporadic abortion group was slightly higher than that in recurrent miscarriage group(63.1%> 61.5%),but there was no significant difference between the two groups(P> 0.05);The abnormal rate of NGS copy number variation in sporadic abortion group was slightly higher than that in recurrent miscarriage group(33.8%>21.2%),but there was no significant difference between the two groups(P> 0.05).Conclusions 1 Embryonic chromosome aberration is the main genetic cause of early spontaneous abortion.2 full chromosome aberration DNA sequencing technology is simple and rapid,accurate and efficient,high-throughput and other characteristics,the application of the technology to detect early spontaneous abortion embryonic villi can find the main cause of early spontaneous abortion,the next pregnancy to provide guidance and genetic counseling.
