Objectives : To observe effects of IL-31 on the expressionof CCL2 in alveolar macrophages in asthmatic mice.Methods:Female BALB/c mice were assigned randomly to asthma group(AS,n=10)and saline control group(NS,n=10).Two groups were respectively sensitized on days 1and 13 by hypodermic injection combined with intraperitioneal of OVA and normal saline,challenged with 10% OVA or normal saline repeatedly for five days on the 19 th day.Mice were sacrificed 24 hours after the final challenge.The bronchoalveolar lavage fluid(BALF)were collected for the total number of cells and classification.Adherent method is used to obtain AMs.Observing cell morphology.Cultured in vitro for 12-24 hours.The cell survival rate was detected with trypan blue dye exclusion test,and cell purity was determined by Wright-Giemsa staining.Cell phenotype was identified by FCM.Mice from AS and NS were killed to obtain AMs.Adding different concentration(10ng/ml、50ng/ml、100ng/ml)IL-31 to AMs.After adding sample 12 h,24h,36 h,by means of PCR to detecting the expression of CCL2 level.CCL2 expression increased with the concentration and time of IL-31 intervention.Results:The total cell count and differential cell counts,the score of airway inflammatory infiltration,airway wall thickness in lung tissue in AS group were significantly higher than those in NS group.AMs morphology presented mixed shapes such as circular,elliptical and spindle shapes.The trypan blue test showed a survival rate≥95 % of the AMs and Wright-Giemsa staining indicated a purity≥ 95 % of the AMs.High-level expression of CD11c、F4/80.The total amount of CCL2 expressed by alveolar macrophages in group AS was higher than that in group NS under the intervention of IL-31.The expression of CCL2 was proportional to the concentration of IL-31.At the same concentration,the expressionof CCL2 was proportional to the duration of IL-31 addition. IL-31 can induce the expression of CCL2 in AMs in asthmatic mice,CCL2 expression i n c r e a s e d w i t h t h e c o n c e n t r a t i o n a n d t i m e o f I L-3 1 i n t e r v e n t i o n.Conclusions: IL-31 can promote the expression of CCL2 in AMs in asthmatic mice,with the increase of time and concentration of IL-31 intervention,CCL2 expression can be proportional to.
