Objective: In this study,we detect Lemur tyrosine kinase 3(LMTK3)expression in the serum and tissues of patients with papillary thyroid carcinoma(PTC),and compare its expression level with patients’ characteristics for the sake of its clinical significance.This research may provide previous dates for discovering a new maker for PTC treatment.Methods:1.The expression of serum LMTK3 and its significance in PTC patients.In this part,postoperative serum samples from PTC patients(n=31),thyroid benign lesion(TBL)(n=32)and Health volunteer(HV)(n=24)were collected.By Enzyme-linked immunosorbent assay(ELISA),the serum LMTK3(s LMTK3)expression was measured in those samples.Meanwhile,we traced the expression levels of serum Thyroglobulin(Tg)detected in hospital.Then,we tried to discover its potential significance in terms of diagnosis compared with serum Tg.2.The expression level of LMTK3 and its significance in PTC tissues.Totally 10 PTC paired cancerous and matched adjacent normal frozen tissue samples,10 TBL frozen tissue samples,106 PTC paraffin-embedded tissues and 22 TBL paraffin-embedded tissues were collected from patients with postoperatively pathological diagnosis.By q RT-PCR(q PCR)and Western blot(WB),we measured the expression of LMTK3 m RNA and protein in those frozen tissues.Then,conduct immunohistochemistry(IHC)on those paraffin sections with LMTK3 antibody and ERα antibody,and investigated their clinical significance.Results:1.The mean concentration of s LMTK3 in PTC group(1.51 ± 1.22)ng/ml was obviously higher than TBL group(0.66 ± 0.87)ng/ml(P<0.05)and HV control group(0.65 ± 0.91)ng/ml(P<0.05).Using receiver operation characteristic curve(ROC curve),we compared s LMTK3 expression of PTC group with control group 1(TBL group)and control group 2(TBL and HV group),and acquired their area under the curve(AUC): 0.720(P=0.006)AND 0.709(P=0.001),respectively.s LMTK3 may be superior to serum Tg in terms of diagnosis of PTC.2.LMTK3 expressed higher in PTC tissues instead of in PTC matched adjacent normal tissues(P<0.001)or TBL tissues(P<0.001).Besides,the expression of LMTK3 positively associated with patient’ clinical TNM stage(P=0.010),however had no correlation with age,gender,tumor size,lymph node metastasis and distant metastasis(P>0.05).3.Estrogen receptor α(ERα)expression increased higher in PTC tissues rather than in PTC matched adjacent normal tissues(P< 0.001)or TBL tissues(P< 0.001).Moreover,the expression of ERα positively correlated with tumor size(P=0.017),However had no correlation with age,gender,lymph node metastasis,distant metastasis and clinical stage(P>0.05).4.According to statistical analysis,the sensitivity and specificity of LMTK3 were significantly higher than ERα in PTC tissues.Additionally,LMTK3 expression had positive correlation with ERα(r=0.292,P<0.001).Conclusion:1.LMTK3 overexpressed in serum of patients with PTC instead of ones with TBL or HVs.The sensitivity and specificity of s LMTK3 was better than serum Tg.2.LMTK3 expression was up-regulated in PTC patients’ tissues,and had the positive association with clinical stage,which suggested the potential roles of LMTK3 in tumorgensis and development of PTC.3.LMTK3 had positive correlation with ERα in PTC tissues,which hint LMTK3 may interact with ERα in the PTC cells.
