| Objective: Cognitive impairment is a common disease in old people,and vascular cognitive impairment(VCI)refers to the cognitive disorder associated with cerebrovascular disease,it can develop from mild cognitive impairment to severe dementia.It is of great significance to improve the quality of life of patients with VCI and to control the incidence of Alzheimer’s disease by exploring new drugs or treatments.This study aimes to explore the effect of donepezil combined fasudil hydrochloride on TNF-α,hs-CRP and IL-6 in serum of patients with VCI.Methods: The 60 patients with VCI hospitalized in the first central hospital of Baoding from January,2016 to December,2016 were selected and randomly divided into two groups by using the method of digital table: the control group and the observation group,each group had 30 cases;the patients in the control group(namely the donepezil treated group)were orally treated with 5 mg donepezil hydrochloride once a day;while the patients in the observation group(namely the donepezil combined fasudil treated group)were treated with fasudil hydrochloride 30 mg via an intravenous drip once a day for 21 days on the basis of oral donepezil.The therapeutic effect was compared between the two groups using the MMSE and the ADAS-cog scores;The levels of TNF-α and IL-6 in serum of patients in both two groups were detected before and after treatment by radioimmunoassay and the levels of hs-CRP in serum of patients in both two groups were detected before and after treatment by immune turbidimetry.Results: Treatment with both donepezil combined fasudil hydrochloride and donepezil hydrochloride alone could improve the cognitive function of the patients with VCI in different levels.Comparing the MMSE and ADAS-cog scores of the two groups patients between pretherapy and post-treatment,the MMSE scores of the two groups patients after treatment increased significantly than that before treatment,while the ADAS-cog scores of the two groups patients after treatment reduced significantly than that before treatment(P<0.05);Comparing the MMSE and ADAS-cog scores between the two groups patients after treatment,the MMSE scores of the donepezil combined fasudil treated group patients were much higher than that of the donepezil treated group patients,while the ADAS-cog scores of the donepezil combined fasudil treated group patients were much lower than that of the donepezil treated group patients(P<0.05).After treatment with donepezil hydrochloride alone,the patients with VCI made significant improvements in the ability of the directional orientation of time and place(P < 0.01),and made improvements in the ability of memory and implement(P<0.05),but made no improvements in the ability of language and computing.However,after treatment with donepezil combined fasudil hydrochloride,the patients with VCI made significant improvements in the ability of implement and the directional orientation of time and place(P<0.01),and made improvements in the ability of language,memory,and computing(P<0.05).Moreover,the patients treated with donepezil combined fasudil hydrochloride made more significant improvements in the ability of the directional orientation of time and place,language,computing,and impairment than those treated with donepezil hydrochloride alone.The levels of TNF-α,hs-CRP and IL-6 in serum of the patients with VCI reduced significantly after treatment compared with pretherapy in each groups(P<0.05);meanwhile,the levels of TNF-α,hs-CRP and IL-6 in serum of the patients treated with donepezil combined fasudil hydrochloride reduced significantly than those of the patients treated with donepezil hydrochloride alone after treatment(P<0.05).Conclusion: Treatment with donepezil combined fasudil hydrochloride makes better improvement in the cognitive function and significant reduction in the inflammatory factors levels in serum of the patients with VCI compared to treatment with donepezil hydrochloride alone.Results suggest that fasudil hydrochloride can improve the cognitive function of the patients with VCI.This study provides more selectivity for clinical treatment of VCI. |
