Purpose: Shiweitiaozhi decoction(It was composed of Zexie,Danshen,Shengshanzha,Jiaogulan,Shuihonghuazi,Chaih,Jianghuang,Shouwuteng,Juemingzi and Chongweizi.)has the function of soothing the liver and regulating Qi,purging turbidity and removing blood stasis,which has been proved significantly effective in treating nonalcoholic fatty liver disease(NAFLD)by years of clinical observation.To further study,the rat model of NAFLD was obtained by high fat diet method and then drug intervention was carried out.We observe the blood lipid,liver function,liver histomorphology and liver tissue PPARαm RNA expression to explore the possible mechanism of the Shiweitiaozhi decoction in treating NAFLD.Methods: Forty healthy male Wistar rats were randomly divided into four groups after being fed adaptively for 1 week: normal group(N),model group(M),Fenofibrate group(X),Shiweitiaozhi decoction group(Z),each group of 10.The rats of normal group were given conventional feed,and the other groups were given high fat diet.After 8 weeks,except for the normal group,one rat was sacrificed from each of the other groups,and the liver histopathological examination was performed to determine whether the model was successful or not.After the model was established successfully,the rats of normal group and model group were given normal saline by intragastric administration,the treatment group were gavaged with medicine.All rats were sacrificed after 4 weeks intervention,serum and liver tissue samples were collected.The contents of triglycerides(TG),total cholesterol(TC),alanine aminotransferase(ALT)and aspartate aminotransferase(AST)in serum were measured by the method of 2,4-dinitrophenylhydrazine.The contents oftriglyceride(TG)and total cholesterol(TC)in liver tissue were measured by the method of Chromatography Visible spectrophotometry.The pathological changes of liver tissue were observed by hematoxylin-eosin staining(HE)staining.The expression levels of PPARα mRNA in rat liver tissue were detected by reverse transcription Polymerase chain reaction.Results:1 The comparison of general condition among diffrient groupsIn the course of the experiment,the rats in each group were not dead.The rats in the normal group were of good mental,physical activity and skin.Their weight was gradually increased.The initial weight of the model group increased significantly,and the rate of growth was faster than that of the normal group.From the age of 7-8 weeks,the weight gain of the rats in model group was slower than that of before.The rats in model group were dispirited,of impaired appetite,messy fur,dark hair color and sticky stool.The rats in the intervention group had the above changes to some extent,but between the normal group and the model group.2 The result of Histopathological examination of liver in ratsIn the normal group,the structure of the liver lobules was complete,the contours were clear.The size of the hepatocytes was consistent and the structure was normal.the lipid-free drops and the vacuoles were infiltrated,and no inflammatory cells infiltrated.The nucleus was normal,and the cytoplasm was rich.In the model group,the structure of the hepatic lobule was disordered,and the arrangement of the hepatic cord was disappeared.The inflammatory cells were infiltrated.Most of the hepatocytes were swollen and the cytoplasm appeared a large number of lipid droplets with different sizes.In Chinese herbal medicine group the structure of liver lobulars was complete,the cells were clear outline,no obvious swelling of liver cells,some cells could be seen a small amount of small lipid droplets,no obvious inflammatory cell infiltration.Western medicine group liver lobular structure slightly disorder,mild swelling of liver cells,cytoplasm can be seen within a small amount of lipid droplets ranging from size,no significant inflammatory cellinfiltration.3 The comparison of serum and liver tissue indexes in rats3.1 The comparison of serum TG andTC among diffrient groupsCompared with the normal group,the levels of serum TG and TC in the model group were significantly higher than those in the normal group(P<0.05).Compared with the model group,the serum TG and TC of the Shiweitiaozhi decoction group and the Fenofibrate group were significantly lower(P <0.05).Compared with the Shiweitiaozhi decoction group,the serum TG and TC of the Fenofibrate group were lower,which was statistically significant(P <0.05).3.2 The comparison of serum ALT andAST among diffrient groupsCompared with the normal group,the serum ALT and AST in the model group were significantly higher(P <0.05).Compared with the model group,the levels of serum ALT and AST were significantly lower in the Shiweitiaozhi decoction group and the Fenofibrate group(P <0.05).Compared with the Fenofibrate group,serum ALT and AST were decreased in the Shiweitiaozhi decoction group,but the difference has no statistical significance(P> 0.05).3.3 The comparison of TG Levels and TC Levels in Liver Tissue among diffrient groupsCompared with the normal group,the levels of TG and TC in the liver tissue of the model group were significantly increased(P <0.05).Compared with the model group,the levels of TG and TC in the liver tissue of the the Shiweitiaozhi decoction group and the Fenofibrate group were significantly lower than those of the model group(P<0.05).Compared with the Fenofibrate group,the levels of TG and TC in the liver tissue of the Shiweitiaozhi decoction group were decreased,and the difference was statistically significant(P <0.05).4 In rat liver tissue PPARαm RNA expression levelCompared with the normal group,the levels of PPARαmRNA in the liver tissue of the model group was significantly decreased(P <0.01).Comparedwith the model group,the levels of PPARαmRNA in liver tissue of Shiweitiaozhi decoction group and the Fenofibrate group were significantly higher(P <0.05).The level of PPARαmRNA in liver tissue of Shiweitiaozhi decoction group was lower than that of Fenofibrate group,but the difference was not statistically significant(P> 0.05).Conclusions:1 Shiweitiaozhi decoction can improve the NAFLD model rats in general condition and liver histomorphological significantly,which is effective prescription of treatmenting non-alcoholic fatty liver disease.2 The levels of TC,TG,AST and ALT in serum and the levels of TC and TG in liver tissue were adjusted to improve the pathological changes of liver lipid metabolism and lipid abnormal deposition in NAFLD.3 The expression level of PPARαmRNA in liver tissue can be up-regulated by Shiweitiaozhi decoction,which may be an important mechanism to improve liver lipid metabolism and reduce the deposition of lipid in hepatocytes to treat nonalcoholic fatty liver disease.
