| Objective : To investigate the biological characteristics of monoclonal antibodies against avian influenza virus(AIV)and provide reference for the study on avian influenza virus infection and pathogenicity.Method:Monoclonal antibodies(m Abs)against AIV H5N1 were prepared and it’s characteristics were identified including subtype,titer,hemagglutination inhibition activity and cross-reactivity with other influenza viruses.Besides,western blot and Immunohistochemical staining methods were conducted to test the combination of antibodies and antigen(H5N1)and human normal tissues.Immunohistochemistry and immunofluorescence were used to further verify the cross reactivity of the monoclonal antibody and human tissue at the cellular level.Results : In this experiment,39 strains Clone antibody were obtained.Immunohistochemical analysis showed that 2 m Abs(H5-32 and H5-35)cross-reacted with human tissues kidney and pancreas respectively.Monoclonal antibody H5-32 coud cross-react with human renal tubular epithelial cells HKC-8.Conclusions:Immunohistochemical analysis showed that 2 m Abs cross-reacted with human tissues kidney and pancreas respectively.This is consistent with autopsy reports of deaths from avian influenza viruses in the past,that in addition to lung injury,myocardial edema,congestion,edema,acute renal tubular necrosis and other organ damage occurred in the patients with avian influenza.These data indicated that there have some association between the AIV H5N1 with human tissues,which may provide reference for the study on avian influenza virus infection and pathogenicity. |
