| ObjectiveInvestigate the protective effect of water-soluble coenzyme Q10 on the apoptosis of Parkinson’s disease(PD)cell model and its possible mechanism,explore the effect of water-soluble CoQ10 on rotenone-induced PC12 cells mitochondria damage protection mechanism for CoQ10,provide a theoretical basis on the treatment of PD.Methods1.PC12 cells(induced by nerve growth factor)were injured by rotenone(Rot),and choose appropriate concentration set PD cell mode.2.Determine the concentration of water-soluble CoQ10 as the therapeutic concentration,the experiment was divided into control group,Rot group,CoQ10 group and CoQ10 treatment group.3.The cell viability was measured by cck-8,observe the cells morphology under inverted microscope at the same time.4.Use the ROS assay kit to detect intracellular ROS changes in each group.5.Use JC-1 mitochondrial membrane potential assay kit and flow cytometry analysis mitochondrial membrane potential(MMP)Change.6.MitoTracker mitochondrial fluorescent probe was labeled with mitochondria,and the mitochondrial morphological changes were observed by confocal laser scanning microscopy.7.Western blotting was used to detect the expression of apoptosis protein and mitochondrial fusion/fission protein.Results1.Cell viability decreased significantly after rotenone treatment 24 h,water-soluble CoQ10 can improve the PC12 cell viability.2.ROS level increased after rotenone treatment 24 h,water-soluble CoQ10 can reduce the level of ROS.3.Mitochondrial membrane potential reduction and mitochondrial fragmentation of PC12 cells induced by rotenone treatment.4.Western blotting experiments show that water-soluble CoQ10 can reduce rotenone induced Caspase-9,active Caspase-3 and Bax expression,increase the expression of Bcl-2 and revent AIF nuclear translocation.;water-soluble CoQ10 can reduce rotenone induced Drp1 expression,increase the expression of Mfn2 and OPA1,there was not significantly effect on the expression of Mfn1 and Fis1.Conclusion1.Water-soluble CoQ10 can stabilize the reduction of MMP and mitochondrial fragmentation in PC12 cells.2.Water-soluble CoQ10 has protective effect on rotenone induced apoptosis in PC12 cells,the mechanism may be through scavenging ROS in cells,decrease caspase-9,active caspase-3 and Bax expression,increase the expression of Bcl-2 and prevent AIF nuclear translocation.3.Water-soluble CoQ10 can improve rotenone-induced mitochondrial fusion/fission disorder in PC12 cells,the mechanism may be through reduce the expression of Drp1,increase the expression of Mfn2 and OPA1. |
