Objective: Breast cancer,ranking the first in morbidity of female common carcinoma,is a malignant tumor.It is reported that the morbidity of breast cancer has increased gradually.Recent researches illustrate that SIRT1(silent information regulator 1)plays an important role in the formation and development of a variety of human cancers.However,there is an argument about the extent to which SIRT1 has an impact on the breast cancer.This study,on the basis of clinical practice,aims to investigate the expression of SIRT1 in operable breast cancer tissues and adjacent tissues,to evaluate its relationship with clinicopathologic features,molecular classification and patient prognosis,and to discuss its role in the formation and development of breast cancer.Methods: This study collected 164 cases of breast cancer tissues and 66 cases of corresponding adjacent tissues,which comes from General Hospital of Ningxia Medical University during the period from January 2009 to January 2012.Each case maintains complete clinical data.To investigate the expression of SIRT1 in breast cancer tissues and corresponding adjacent tissues,the tissue microarray technology and immunohistochemistry staining technique were applied,and a statistical analysis was developed in this study.Results: Based on gene chips of 164 cases of breast cancer tissues and 66 cases of corresponding adjacent tissues,applying immunohistochemistry staining technique,this study found that the expression of SIRT1 in human breast cancer tissues(67.7%,111/164)is much higher than that in adjacent tissues(13.6%,9/66),having a statistical significance(P<0.001).The expression of SIRT1 in breast cancer tissues is correlated with histological grade and axillary lymph nodes metastasis(P<0.05).The expression of SIRT1 is also related to PR,Ki-67(P<0.05).In human breast cancer tissues,patients with positive expression of SIRT1 maintain a much lower diseases free survival and overall survival than those with negative expression of SIRT1,having a statistical significance(P<0.05).The Cox regression analysis indicates that SIRT1 is an independent prognostic factor for 5-year diseases free survival and overall survival of breast cancer(P<0.05).Conclusion: The expression of SIRT1 in human breast cancer tissues maintains a much higher level than that in adjacent tissues,which indicates that SIRT1 may promote the formation and development of breast cancer.The higher histological grade is and the more axillary lymph nodes metastasis has,the higher expression of SIRT1 is.In addition,SIRT1 is positively correlated with PR,Ki-67.In different molecular subtypes of breast cancer,Luminal breast cancer keeps a high positive expression of SIRT1.Breast cancer patients with high expression of SIRT1 remain poor prognosis and poor survival rate.The expression of SIRT1 is an independent risk factor for the prognosis of patients with breast cancer.
