Objective:The study aimed to investigate the relationship of plasma miR-27b and miR-10b with cerebral atherosclerosis and abnormal lipid metabolism.Methods:A total of 47 healthy subjects,38 patients with atherosclerosis,52 patients with large-artery atherosclerosis,and 52 patients with small-artery occlusion were enrolled,and levels of plasma miR-27b and miR-10b were determined by real-time fluorescence quantitative PCR.To evaluate the extent of cerebral artery atherosclerosis,the control group was set up as non atherosclerosis group,LAA group and AS group were set up as atherosclerosis group.The atherosclerosis group was divided into 3 subgroups according to the single,double and multiple branches(more than three and above).To analyze the correlation between the expression of miR-27b and miR-10b in plasma and the extent of cerebral atherosclerosis by Spearman correlation analysis.To analyze the correlation between plasma miR-27b and miR-10b levels and serum lipids by Pearson correlation analysis.To evaluate the accuracy of miR-27b and miR-10b in cerebral atherosclerosis by the receiveroperating characteristic(ROC)curve.Results:The levels of miR-27b,miR-10b and TC in atherosclerosis group were significantly higher than those in control group and SAO group(P<0.05).There was no significant difference between the control group and SAO group(P>0.05),AS group and LAA group(P>0.05).It is suggested that miR-27b and miR-10b are related to cerebral atherosclerosis,and the effect of infarction is less.Spearman correlation analysis showed that the expression levels of miR-27b and miR-10b were correlated with the degree of cerebrovascular atherosclerosis(r=-0.571,P<0.001;r=-0.521,P<0.001).Pearson correlation analysis revealed that the expression level of miR-27b and miR-10b in plasma in atherosclerosis group and non atherosclerosis groups had correlation with the level of TC.The recognition performance evaluation of miR-27b and miR-10b on cerebral atherosclerosis,the area under the ROC curve were 0.932(95%CI:0.884-0.980,P<0.001);0.924(95%CI:,0.870-0.979,P<0.001),the cut-off value is 5.48;5.82,the sensitivity and specificity were 89.4%,91.1%,85.1%,96.7%.Conclusions:The plasma levels of miR-27b and miR-10b in the AS group and LAA group were significantly higher than those in the control group and the SAO group,but there was no significant difference between the AS group and the LAA group,the control group and the SAO group.Moreover,the expression levels of miR-27b and miR-10b in plasma were significantly correlated with the degree of atherosclerosis.This shows that miR-27b and miR-10b are not only associated with cerebral atherosclerosis,but also the higher the expression of miR-27b and miR-10b in plasma,the more extensive the pathological changes of cerebral atherosclerosis.
