Objective To explore the relationship between the expression of TSHR,TSHR-m RN A and the BRAFV600E mutation in patient's tissue and peripheral blood with papillary thyroid carcinoma.Methods Collect 122 patients with papillary thyroid cancer and take papillary thyroid cancer tissues and normal concrol group thyroid tissues as experimental samples during the operation.All of the patients carried out postoperative routine BRAFV600E mutation testing.We measured the expression of TSHR protein in all papillary thyroid carcinoma patients' tumor tissues by immunohistochemical technique.Detect the differences in the expression of TSHR between papillary thyroid cancer(BRAFV600E mutation type 20 cases VS BRAFV600E genotype of wild 20 cases)and normal control group thyroid tissues?20 cases?by western-blot.We applied RT-PCR to test the relative expression of TSHR-m RNA between papillary thyroid cancer tissues(BRAFV600E mutation type 10 cases VS BRAFV600E genotype of wild 10 cases)and normal control group thyroid tissues?10 cases?.70 patients with papillary thyroid cancer,34 patients with benign thyroid disease and 11 healthy volunteers were recruited and their peripheral blood samples were collected.We measured the relative expression of TSHR-m RNA from all of the participants.SPSS17.0 statistical software was app lied to analyze the relationship between the expression of TSHR and BRAFV600E mutation.Results The expression of TSHR protein and TSHR-m RNA in papillary thyroid cancer tissues was significant decreased.The expression level of TSHR protein and TSHR-m RNA in patients with thyroid papillary carcinoma and BRAFV600E mutation,less than those who haven't BRAFV600E mutation.The expression level of preoperative peripheral blood TSHR-m RNA in papillary thyroid carcinoma patients was significantly higher than that in benign thyroid disease patients and healthy volunteers and the expression of TSHR-m RNA fall to normal level absolutely at post-operative 1 day.The expression level of preoperative peripheral blood TSHR-m RN A in papillary thyroid carcinoma patients with BRAFV600E mutation was significantly higher than that in papillary thyroid carcinoma patients without BRAFV600E mutation.Conclusion The expression of TSHR in papillary thyroid carcinoma was influned by BRAFV600E mutation and the expression level of TSHR protein and TSHR-m RN A decreased in papillary thyroid carcinoma tissues with BRAFV600E mutation;As a maker of circulating tumor in patients with papillary thyroid carcinoma,TSHR release peripheral blood associated with BRAFV600E mutation.However,BRAFV600E mutation is not the only cause and initiation factor of the expression of TSHR protein and TSHR-m RNA in papillary thyroid carcinoma.
