Association Between TLR4 Gene Polymorphisms And The Large-artery Atherosclerosis Stroke

Objective To explore the association of TLR4 gene polymorphisms(rs1927911,rs1927914 and rs2149356)and the Large-artery atherosclerosis(LAA)stroke in an ethnic Han population from Qingdao Region.Methods Subjects of ischemic stroke patients were in the Second Affiliated Hospital of Qingdao University Medical College and the same period in our hospital physical examination center physical examination.A total of 105 patients with LAA type cerebral infarction were selected according to TOAST classification,and 128 healthy subjects as controls.After admission,all subjects were collected 3 ml of forearm venous blood by fasting the morning of the next morning for the detection of biochemical indexes and the extraction of leukocyte DNA.All subjects were collected for gender,age,smoking history,history of alcohol consumption,history of hypertension,history of diabetes and history of coronary heart disease.Collect and record data.The polymorphism of rs1927911 and rs1927914 loci in TLR4 gene was detected by polymerase chain reaction-restriction fragment length polymorphism.The polymorphism of rs2149356 locus in TLR4 gene was detected by direct sequencing.SPSS17.0 software was used for statistical analysis.The measurement data of the two groups were compared with the t-test.The metrological data of the normal distribution were expressed by the mean ± standard deviation(?).Count data is applied to X~2 test.The correlation between the influencing factors and the cerebral infarction was analyzed by multivariate logistic regression analysis.p<0.05 was statistically significant;with Hardy-Weinberg equilibrium test sample population representation,p>0.05 with population representation.Results Rs1927911 genotype frequency in LAA group and the control group the difference was statistically significant(p=0.041),rs1927911 allele frequency in LAA group and the control group the difference was statistically significant(p=0.754),rs2149356 genotype frequency and allele frequency in LAA group and the control group the difference was statistically significant(p=0.002,p=0.006),There was no statistically significant difference in genotype frequency and allele frequency between rs1927914 locus between the two groups(p=0.472,p=0.956).Conclusion The polymorphism of rs1927911 and rs2149356 loci of TLR4 gene were correlated with the Large-artery atherosclerotic stroke in Chinese Han population in Qingdao Region,while rs1927914 polymorphism was not associated with LAA type cerebral infarction.The allele A at the rs2149356 locus may be a susceptible gene for LAA cerebral infarction.
