Objective:To investigate the expression of steroidogenic factor-1(SF-1)in the endometrium of early pregnant mice and effect of SF-1 expression on proliferation of endometrial stromal cells and its regulatory mechanism.Methods:Established of mouse pregnancy,pseudopregnancy,artificially induced decidualization model and collected of materials;the expression of SF-1 was detected by immunohistochemistry and Western Blot in the endometrium of early pregnant mice;the primary mouse endometrial stromal cells were isolated and identified;the expression plasmid of pCMV6-SF-1 was transfected by Lipofectamine 2000,PI3 K signaling pathway inhibitor LY294002 and mTOR signal pathway inhibitor Rapamycin and other different ways to deal with the mouse primary stromal cells.Results:Immunohistochemistry and Western Blot showed that the expression of SF-1 was higher in implantation site than in interimplantation sites on the first day of 5,6,8 in the endometrium of pregnant mice;there was no significant difference in the expression of SF-1 on PD1-PD8 in the pseudopregnant mice endometrium;the expression of SF-1 in artificially induced decidualization of mice was significantly higher than that of control group.Overexpression of SF-1 can significantly induce the protein expression of the proliferation related genes PH3 and PCNA and cell proliferation;the results of EdU also showed that overexpression of SF-1 could induce the proliferation;the expression of PI3K-AKT-mTOR signaling pathway related proteins was detected by Western Blot;the results showed that PI3K-AKT-mTOR signaling pathway was involved in the cell proliferation by overexpression of SF-1.Conclusions:1.SF-1 can induce the proliferation of endometrial stromal cells in early pregnant mice.2.Up-regulation of SF-1 expression can induce the proliferation of stromal cells by promoting the activation of PI3K-AKT-mTOR signaling pathway. |
