| Objective:To investigate the effect of emodin on the expressions of ERK1/2 and p38 MAPK in RA Fibroblast-like Synoviocytes,and to study the mechanism of emodin for treating rheumatoidarthritis(RA)Methods:The cells MH7 A culture invitro were divided into five groups:control group,model(TNF-a10ng/ml)group,low,medium and high emodin dose groups.The expression levels of ERK1/2 and p38 MAPK were evaluated by immunofiuorsecence,western blot and RT-PCR methods.Results:Immunofiuorsecence results demonstrated that TNF-a treatment showed a large red fluorescence expression in the nucleus of RAFLS cells.After the treatment with 60μM concentration emodin,the red fluorescence in the nucleus decreased gradually,and then transferred to the cytoplasm;western blot results displayed that the expressions of ERK1/2 and the expressions of p38 MAPK increased significantly in the model group(P<0.05);PCR results displayed that the expressions of ERK1/2 mRNA and the expressions of p38 MAPK mRNA increased significantly in the model group(P<0.05);while compared with the model group,western blot and RT-PCR results displayed that the expressions of ERK1/2 and p38 MAPK in treated groups were significantly reduce compared with model group(P < 0.05),emodin in all dose groups,the expression of ERK1/2,p38 MAPK and mRNA were significantly reduced(P<0.05),and the higher the concentration decreased more obviously(P < 0.05),in a dose dependent manner.Conclusion:(1)TNF-a increaseed the expressions of ERK1/2 and p38 MAPK in Fibroblast-like Synoviocytes is one of the reason of rheuatoid arthritis.(2)Emodin suppressed the expressions of ERK1/2 and p38 MAPK and maybe dose dependent in Fibroblast-like Synoviocytes is one of the mechanism for treating rheuatoid arthritis. |
