Objective:To prove the existence of Caspase-3 gene in normal skin fibroblasts and keloid fibroblasts from the protein level.To study the effect of 5-FU(5-fluorouracil)concentration gradient on the expression of Caspase-3 protein and proliferation,migration and invasion of fibroblasts from keliod.Methods:(1)Immunohistochemistry and Western blot was used to detect Caspase-3 protein expression in keloid and normal skin tissue,keloid fibroblast(KF)and normal skin fibroblasts(NSF);(2)5-FU(5-fluorouracil)concentration gradient effect of the keloid fibroblasts(KF).Western blot was used to detect the expression of Caspase-3 protein expression.CCK-8 test measurement KF cells proliferation.Trans-wells chamber assay KF cells in migration and invasion ability.Results:The keloid tissue,normal skin tissue,KF,NSF have the expression of Caspase-3 protein.The expression levels of Caspase-3 protein were significant differences between KF and NSF(P<0.05);The expression levels of Caspase-3 protein were significantly up-regulated after 2 hours of 5-FU(0.1mmol/L,0.01mmol/L,0.001mmol/L)act on KF and the effects were concentration dependent(P < 0.05);CCK-8 results showed that 5-Fu(0.1mmol/L,0.01mmol/L,0.001mmol/L)inhibited proliferative activity of KF,and the effects were concentration dependent(P<0.05).Trans-well results that 5-Fu(0.1mmol/L,0.01mmol/L,0.001mmol/L)inhibited the migration and invasion activity of KF,and the effects were concentration dependen(P<0.05).Conclusion:Caspase-3 is involved in the regulation of 5-FU induced KF apoptosis.5-FU promotes Caspase-3 activity and inhibits the tumor characteristics such as KF proliferation,invasion,migration and so on.
