Pannonibacter phragmitetus is identified as a facultative aerobic,nonfermentative Gramnegative bacillus.The present study onP.phragmitetus is mainly focused on itspotential in detoxification of heavy metals.P.phragmitetus is an opportunistic pathogen.Although only four cases were reported until now,butthat the number of undetected infections may be large.P.phragmitetusis a multi-drug resistant bacteria.Its antibiotics resistance mechanism,pathogenesismechanism and heavy metal metabolism mechanism ingenomic level have not been reported before.For clarification of these issues,thisP.phragmitetus31801 isolated from a liver abscess patient was performed on antibiotic susceptibility testing(AST)and genome sequencing.This AST results showed that P.phragmitetus 31801 was resistant to aminoglycosides,fluoroquinolones,tetracyclines,nitrofuran antibiotics,and this can guide clinical treatment.P.phragmitetus31801 whole genome sequencing done by PacBio RS II platform,contained a complete chromosome and a closed loop plasmid(p.p-1).It was assinged to P.phragmitetus sp by average nucleotide identity(ANI)analysis.With CRISPRFinder,PHAST,IslandViewer,Mauve and comparative bioinformatics analysis,we found that P.phragmitetus 31801 has 5CRISPRs,3 prophage regions and 21 Gene Islands,and the prophage A was predicted to be horizontallytransferredfrom other bacteria in environment.With ARDB,CARD,RAST,VFDB databases,large number of genes involved in the mechanisms of antibiotics resistance,pathogenesis and heavy metal metabolism were found.For antibiotics resistance genes,we found that β-lactam,quinolones,fosfomycin and other relatedantibiotics resistance genes in P.phragmitetus 31801.From the genomic level,the mechanism of the resistance of this bacterium to β-lactam,quinolones and sulfonamides is explained.For the pathogenesis mechanism,P.phragmitetus 31801 is found to containmany genes encoding for virulence factors,flagellates,secretion systems.For the mechanism of heavy metal metabolism,many genes involved in metabolism of chromium,mercury,cadmium and other heavy metal were identified,all these can explain the potential of P.phragmitetusin the metabolism of heavy metals.The above findings elucidated the capacity of P.phragmitetus 31801 antibiotic resistance,pathogenicity and heavy metal metabolism at the genomic level,laying the foundation for further laboratory research. |
