Deguelin Inhibited Cell Growth By PI3K/Akt Pathway In Mantle Cell Lymphoma

Objects To investigate the effect of deguelin on cell proliferation and apoptosis in mantle cell lymphoma cell lines Jeko-1 and Mino and its possible mechanism with the PI3K/Akt pathway.Methods The proliferation of Jeko-1 and Mino cells after exposure to indicated concentration of deguelin were determined by MTT.Cell apoptosis was checeked by flow cytometry.The protein of Bcl-2,Bax,Caspase-3,Cyclin D1,and protein Akt,p-Akt,p-GSK-3β were detected by Western Blot.Results Cell proliferation was inhibited by deguelin in Jeko-1 and Mino cells lines.In Jeko-1 cells,growth rate was(83±2.33)%,(58±3.45)%,(42±3.12)% after exposure to deguelin in 20,40,80nmol/L for 24h(p<0.05),cells proliferation rate was(83±2.33)%,(75±3.72)%,(52±3.56)% after exposure to deguelin in 20nmol/L for 24,48,72h(p<0.05).In Mino cells,cell growth rate was(86±2.79)%,(52±3.02)%,(36±3.77)% after exposure to deguelin in 20,40,80nmol/L for 24h(p<0.05),cells proliferation rate was(86±2.79)%,(70±3.85)%,(62±3.71)% after exposure to deguelin in 20nmol/L for 24,48,72h(p<0.05).After 24 h exposure to deguelin in Jeko-1 cells with 20,40,80nmol/L,the apoptosis rate was(13.74±2.82)%,(32.19±3.17)%,(46.93±3.68)%(p<0.05).The apoptosis rate of Mino cells was(11.33±2.46)%,(30.52±3.82)%,(55.72±3.62)% after 24 h exposure to deguelin with 20,40,80nmol/L(p<0.05).Deguelin significantly down-regulated Cyclin D1 and Bcl-2,and up-regulated Bax and Caspase-3(p<0.05).And Deguelin significantly down-regulated p-Akt and p-GSK-3β(p<0.05),whereas there was no alteration in Akt.Conclusions Deguelin inhibits cells proliferation and induces apoptosis in Jeko-1 and Mino cells.It down-regulates Cyclin D1,Bcl-2 and up-regulates Bax and Caspase-3.Deguelin dephosphorylates PI3K/Akt pathway,resulting down-regulation of p-Akt and p-GSK-3β.
