Objective: To investigate the genetic polymorphism and its Forensic application of 15 STR loci from Amp FLSTR IdentifilerTM system in personal identification and paternity testing in Han race population in Xiamen Fujian.Collect other domestic provinces and cities population gene frequency data,calculating the genetic distance and then build the phylogenetic tree,the genetic relationship between studies.Methods:(1)Using Amp FLSTR IdentifilerTM fluorescent compound amplification system kit,randomly selected from 400 Han unrelated individuals in Xiamen of Fujian as the research object,PCR amplification technique,capillary electrophoresis and fluorescence detection technology to separate and genotype.The heterozygosis(H),power of discrimination(PD),polymorphism information content(PIC),probability of paternity exclusion(PE)and matching Probability(PM)were calculated by Modified-powerstate software.The exact test of the Hardy-Weinberg equilibrium was also performed According to the acquired xiamen people and other ethnic groups of periodical paper(including other city in the province and outside the province of Fujian province)gene frequency distribution data,application arlequin3.5.2.2 version of the software in the population 15 STR Compairisons function module analysis comparing the crowd the differences of allele frequency distribution.(2)Literature collected a total of nine groups of nine forensic medicine commonly used STR loci gene frequency,genetic distance was calculated by the statistical software package POPTREE.Neighbor-joining method to build the phylogenetic tree,research of all ethnic groups,the genetic relationship between different people.(3)Through the public databasesor peer-reviewedjournals,STR loci frequency data of other ethnic groups was collected from all provinces in China.After gathered the data from this study and references,we calculated the frequencies of 9 commonly used forensic STR loci in China’s 9 ethnic groups.The genetic distance was calculated by the POPTREE statistical package,and the phylogenetic trees was constructed by the Neighbor-joining method t O study the genetic relationships between the various ethnic Groups.Results:(1)Genotype distribution of 15 STR loci are accord with Hardy Weinberg equilibrium(P > 0.05).15 STR genetic markers are highly polymorphic,heterozygosity between 0.580 0.580,matching probability of o.036 0.148,the individual discrimination is 0.798 0.967,polymorphism information content was 0.560 0.850,the father ruled out rate was 0.268 0.730.Except CSFIPO(0.886),D3S1358(0.886),TH01(0.852),TPOX(0.798)DP were greater than 0.9,heterozygosity were greater than 0.7,the PIC were greater than 0.7.(2)15 loci:1 D18S51,v WA,D21S11,D7S820,FGA,TH01,D13S317,this eight D19S433 loci in Xiamen and Fuzhou,Longyan,Quanzhou gene frequency distribution in the comparison have statistical significance(P < 0.05);2Made in comparison with the provincial,15 loci was statistically difference(P < 0.05),among them,the loci D2S1338,D21S11,v WA,D7S820 and D13S317 and provincial group were statistically significant(P < 0.05).(3)Phylogenetic tree was constructed using the UPGMA method based on genetic distances of 9 populations.In the phylogenetic tree,the Xiamen Hans is the nearest to Guangdong Hans,and then comenear to Hans in Xuancheng Zhejiang,to Yi Sichuan,to Zhoushan Hans,to the kazak in Xinjiang and,Lanzhou Hui polymerization respectively,then the han polymerization with Beijing,next to Gansu Dongxiang phase polymerization.Conclution:(1)All loci in the Han population in Xiamen has high genetic polymorphism,genetic polymorphism,genotype frequency distribution has certain difference,so the IdentifilerTM system 15 STR loci have higher forensic individual identification and paternity tests use value;IdentifilerTM detection system of 15 STR loci is not completely conform to the Han nationality population in Xiamen,Fujian province,and even China group of individual identification and triplet paternity testing requirements,therefore,the researchers need to seek further more area high identification capability of STR loci.(2)1D18S51,v WA,D21S11,D7S820,FGA,TH01,D13S317,D19S433 the eight loci in xiamen and fuzhou,longyan,quanzhou has larger significance in the study of genetic differences;2Longitudinal loci D2S1338,D21S11,v WA,D7S820 and D13S317 can be applied to other groups outside of xiamen and fujian province to study the genetic differences;3 v WA,D21S11,D7S820 and D13S317 these four loci in xiamen and fujian province,the study of genetic diversity in the province has great application value;(4)Each ethnic group genetic background both particularity and independence,also have mingled.(3)Build system of domestic nine groups evolutionary tree,found that the han population in xiamen in fujian province and guangdong han nationality population genetic distance closer;More obvious difference between various nationalities in different regions,the differences between various nationalities is more obvious. |
