A Correlation Study Between Quantitative DKI Parameters And Immunohistochemical Labeling Indices In Nasopharyngeal Carcinoma

Objective: To evaluate the expression of immunohistochemical labeling indices which reflect tumor water diffusion(cell density,CD),tumor angiogenesis(microvessel density,MVD),tumor proliferation(nuclear proliferation index,Ki67)and vasopermeability(aquaporin-1,APQ-1)in nasopharyngeal carcinoma(NPC),and explore the correlation between above pathological indices.The most important is to study the correlation between the diffusion kurtosis imaging(DKI)quantitative parameters and pathological indicators in NPC.Materials and Methods: sixty-six patients(2013.09-2014.12,Fujian Provincial Cancer Hospital)with NPC received DKI and nasopharyngeal biopsy before treatment.We performed MRI examinations on a 3.0 tesla MR scanner system and investigated quantitative parameters(MD and MK)by using DKI sequence.CD expression was deteted by HE staining.MVD,Ki67 and AQP-1 expression were detected by immunohistochemistry(IHC)staining.Finally,the correlations were analyzed by Spearman's rank coefficient analysis.A P value<0.05 was considered statistically significant.Results: We found that the positive expression rate of Ki67 was 93.3%,AQP-1 was95.4%,CD and MVD were 100% in these NPC cases.CD expression was significantly correlated with Ki67(?=0.377,P=0.003),and MVD expression was significantly correlated with AQP-1 expression(?=0.403,P=0.001).CD and Ki67 expression had no correlation with MVD and AQP-1 expression.MD was negatively associated with CD and Ki67 respectively(r=-0.357,P=0.004;r=-0.412,P=0.001).MD had no correlation with MVD and AQP-1 expression.There were no significant relationship between the MK and any immunohistochemical labeling indices in our study.Conclusion: DKI quantitative parameter of NPC(MD)has negative correlation with the CD and Ki67,which provide pathological mechanism for DKI to predict the prognosis of nasopharyngeal carcinoma after radiotherapy.
