ObjectiveTo investigate the mechanism underlying brain injury-induced promotion of peripheral nerve repair by comparing it with the effects of tacrolimus,an immunosuppressant with well-established neuroregenerative properties,in rat models of sciatic nerve injury.MethodsAll animals underwent sciatic nerve transection,and were randomly assigned to four groups to receive tacrolimus or cerebral contusion: A1(brain injury + tacrolimus);A2(brain injury alone);B1(tacrolimus);B2(control:sciatic nerve transection alone),2weeks.At 4,8 and 12 weeks after surgery,we measured sciatic functional index and gastrocnemius muscle wet weight,and examined collagen formation and morphology of muscle and nerve fibers using Masson’s trichrome,hematoxylin-eosin,and toluidine blue staining.At8 and 12 weeks after surgery,we also measured action potential amplitude in the sciatic nerve trunk,and conducted horseradish peroxidase tracing.ResultsTacrolimus combined with brain injury promotes peripheral nerve repair more than brain injury or tacrolimus alone.ConclusionThe mechanisms underlying brain injury-induced promotion of peripheral nerve repair are different from those of tacrolimus,and require further investigation.
