| Objective: To explore the mechanism of Huazhuo Xiaozheng Ruangan Granules protect the immune organs and treat liver cancer,observe the effect of Huazhuo Xiaozheng Ruangan Granules inhibiting H22 transplanted hepatocellular carcinoma growth and the expressions of CyclinD1 and p16.Methods: After injecting H22 hepatoma cell into their right axila,mice were randomly and equally divided into four groups: model group(M),Huazhuo Xiaozheng Ruangan Granules group(Z),5-Fu group(X),and Huazhuo Xiaozheng Ruangan Granules combination with 5-Fu guoup(C),10 mice in each group.During 10 days,durgs were given to mice,the M group received treatment of double distilled water and normal saline;the Z group treated by Huazhuo Xiaozheng Ruangan Granules and normal saline;the X group treated by double distilled water and 5-Fu;the C group received both drugs.We will get the general health condition,inhibiting rate,immune organs index and q value.We will also observe the morphological changes of the tumor tissue.CyclinD1 and p16 are examined by using immumohistochemistry straining method in the tumor tissue.Results:1 General condition of the mice: There was no death during treatment after built the model successfully.Z group and C group were better than X group and M group in mental state,foraging activities,action sensitivity;2 Inhibitted function on H22 tumor cell:inhibiting rate was calculated after death,peeling tumor tissue wholely.The tumor inhibiting rate of Z group was 33.6%,while the X group inhibiting rate was 40.6%.At the same time,the C group inhibiting rate raised up to 54.3% exceeding the standard of curative effect.It was proved that drugs were effective.Inhibiting rate of the C group compared with the Z group increased by 38.1%,and increased by 25.2%compared with the X group.It was say that both drugs used was better than single;3 Calculated by weighting the spleen,thymus and body.The Z group compared with the M group,both the spleen index and the thymus index improved,and the difference was statistically significant(P<0.01).The Z group compared with the X group and the C group,the immune organ index both increaced,the difference were statistically significant(P<0.01).The C group compared with the X group,the thymus index increased,the difference was statistically significant(P<0.01),while the spleen index increased,the difference was not statistically significant(P>0.05).It was conclusion that traditional Chinese medicine enhanced immunity and protected the immune organ,and that 5-Fu damage to the immune organ;4 Tumor cell grew in the M group by diffuse or crumb,and cells were closely arranged,large nuclear,less cytoplasm,obviously specificity.In the Z group,the tumor cells died more than the M group.However,loosely arranged cells,a large number of dead cells were founded in the X group,and the cells decreased.The cells were the worst in the C group;5 The expressions of CyclinD1 were the most in the M group,and the expressions of CyclinD1 were the least in the C group.The Z group decreased by 26.7%,the X group decreased by 44.5%,the C group decreased by 52.5%compared with the M group,and the difference was statistically significant(P<0.01).The expressions of CyclinD1 in the C group decreased and obviously diferent from the Z group and the X group(P<0.01).However,the expressions of p16 were negative in the M group,and the expressions of p16 were the most in the C group.Expressions of p16 in H22 tumor cells increased in treatment groups compared with the M group(P<0.01).The expressions of p16 in the C group were more than the Z group and X group.Conclusion:1 Huazhuo Xiaozheng Ruangan Granules can inhibit H22 transplanted hepatocellular carcinoma growth,the reason why is might be related with the down-regulation of CyclinD1 expression and the up-regulation of p16 expression.2 Huazhuo Xiaozheng Ruangan Granules combined with 5-Fu not only has synergistic antineoplasmic activity,but also reduces the damage caused by chemotherapy of 5-Fu. |
