Objectives: By observing and studying the effect of Xiang Lian Hua Zhuo Jie Du Decoction for Ulcerative Colitis(Ulcerative Colitis,UC)model rats on general conditions,disease activity index(DAI)score,pathological histology and the expression of TLR4/My D88/NF-?B signal pathway(TLR4,My D88,NF-?B)m RNA in the colon tissue,the purpose of the study was to explore the role of TLR4/My D88/NF-?B signal pathway in the pathogenesis of UC.Combining the traditional medicine and modern experiment technology,providing theoretical and experimental basis in the clinical application and promoting.Methods: 60 healthy male wistar rats were adaptability fed to 200-250-g/a and then they were divided into 6 groups randomly:the normal group(NG),the model group(MG),the Srlfasalazine group(SASP),the high-dose group(HDG),the middle-dose group(MDG)and the low-dose group(LDG).Each group included 10 rats.Besides NG,the other five groups adopted 2,4,6-trinitrobenzene sulfonic acid/ethanol(TNBS/ethanol)to prepare UC rats model.From the 4 days after the building,NG rats in the normal feeding,saline lavage to MG rats,SASP rats using Srlfasalazine Mixed suspension lavage,the Chinese medicine treatment groups used the corresponding drug lavage.Observed and recorded the state of mind,drinking and activity,excrement characters,and so on and so forth of rats in each group every day.DAI scores of each group were recorded and calculated.To fill the stomach after 2 weeks,observed the general view of the colonic mucosa and histopathological changes after all rats were sacrificed.Measured the expression of TLR4,My D88 and NF-?B m RNA by RT-PCR method.Results:1 The general conditions and DAI score of ratsThe general conditions: Rats in NG had responsive fast,good appetite,normal weight growth,good mental state,fur luster.Their faeces were granular.Compared with NG,the rest of the 5 groups of rats gradually appeared listlessness,disheveled and matt fur,reduced activity and diet,weight loss,loose stool or even mucobloody stool.After 14 days treatment,the general conditions of HDG were similar to NG,the improve in MDG and SASP were no obvious compared with HDG,but the two groups were no apparent difference,the improve in LDG were worse than MDG and SASP,but better than MG.DAI score: The score of NG was 0.The score of MG(2.458±0.434)was highest.The score of MG significantly higher than the score of HDG(0.370±0.351),MDG(0.767±0.316),LDG(1.625±0.375)and SASP(0.792±0.305).Each group differences were statistically significant(P<0.05).The score of HDG significantly lower than the score of MDG,LDG and SASP.The differences were statistically significant(P<0.05).The score of LDG higher than the score of MDG and SASP.The differences were statistically significant(P<0.05).There were no statistically significant difference(P>0.05)between MDG and SASP.2 The general observation and histopathological observationThe MG was the worst.The HDG was closely to the NG.There was no significant difference between SASP and MDG,which were better than LDG while worse than HDG.Histopathological observation: NG was normal colonic mucosa of rats.MG was the most serious.IT has been formed ulcer and erosion,It had obvious congestion and edema,and a large number of inflammatory cells infiltration.Its gland disorder was lacked and structured.After the treatment,recoveried colonic mucosa in HDG was similariy to NG,which had no ulcer and erosion,had no obvious congestion,edema,inflammatory cells infiltration,had basic integrity of epithelial cells,and had complete and neat rows of glands.Recoveried colonic mucosa in MDG was similariy to SASP.Theywere better than LDG,but worse than HDG.3 Comparison of TLR4?My D88?NF-?B m RNA expression.TLR4: Compared with NG(0.219±0.038),the expression of MG(0.676±0.036)were increased significantly.The differences were statistically significant(P<0.05).Compared with MG,the expression of other treatment groups were reduced significantly.The differences were statistically significant(P<0.05).the expression of HDG(0.343±0.032)was lower than MDG(0.427±0.030),LDG(0.524±0.034)and SASP(0.419±0.024).The differences were statistically significant(P<0.05).There was no significant difference(P>0.05)between MDG and SASP.The expression of LDG highter than MDG and SASP.The differences were statistically significant(P<0.05).My D88: Compared with NG(0.245±0.030),the expression of MG(0.627±0.021)were increased significantly.The differences were statistically significant(P<0.05).Compared with MG,the expression of other treatment groups were reduced significantly.The differences were statistically significant(P<0.05).the expression of HDG(0.387±0.040)was lower than MDG(0.540±0.025),LDG(0.542±0.036)and SASP(0.496±0.035).The differences were statistically significant(P<0.05).The expression of MDG and LDG highter than SASP.The differences were statistically significant(P<0.05).There was no significant difference(P>0.05)between MDG and LDG.NF-?B: Compared with NG(0.244±0.015),the expression of MG(0.624±0.017)were increased significantly.The differences were statistically significant(P<0.05).Compared with MG,the expression of other treatment groups were reduced significantly.The differences were statistically significant(P<0.05).the expression of HDG(0.336±0.019)was lower than MDG(0.479±0.021),LDG(0.516±0.012)and SASP(0.436±0.013).The differences were statistically significant(P<0.05).The expression of SASP lower than MDG.The differences were statistically significant(P<0.05).The expression of MDG lower than LDG.The differences were statistically significant(P<0.05).Conclusion:1 Xiang Lian Hua Zhuo Jie Du Decoction can improve the general state of UC rats including abdominal pain,diarrhea,stool,and so on.The decoction can reduce colon mucosa pathological damage and inhibit the development of inflammation.It also can promote the restoration of colonic mucosa.And there is a dose-effect relationship between its curative effect and dosage.2 The mechanism of action of Xiang Lian Hua Zhuo Jie Du Decoction to treat UC is probably likely to reduce the expression of TLR4,My D88,NF-?B,and inhibit the activation of TLR4/My D88/NF-?B pathway to reduce the release and transcription of inflammatory factors.The aim is to restore intestinal immune balance and promote the healing of ulcer.
