Objective: Detect the difference of serum metabolite in Hepatic alveolar echinococcosis(HAE)patients and healthy people by using ~1H NMR,then analyze the characteristics and differences of ~1H NMR metabolic spectrum,discuss the value of ~1H NMR metabonomics in clinical diagnosis of HAE.Method: The serum of 17 patients with HAE diagnosed in the Department of hepatobiliary and pancreatic surgery,Affiliated Hospital of Qinghai University,and 15 normal persons were collected randomly from July 2016 to September.Detect the Serum metabolites in HAE group and control group by ~1H NMR.PCA,PLS-DA and OPLS-DA were used to pattern recognition of ~1H NMR spectral data,then analyze the difference of metabolites in two groups.Results: Multivariate statistical analysis based on ~1H NMR metabonomics,analysis based on PCA from the scoring charts for distinguishing HAE from healthy people labeled overlapping number two group distinguish sensitivity was 88.23%,the false positive rate was 20%;while PLS-DA and OPLS-DA score plot can significantly discriminate HAE patients and healthy people 100% markers to distinguish,the diagnostic sensitivity of HAE 100% the serum antibody of echinococcosis Ig G diagnostic sensitivity was 76%,significantly reduced the false positive rate of occurrence.PCA,PLS-DA,OPLS-DA analysis results can be used for the diagnosis of HAE,may be better than that of serum antibody of echinococcosis Ig G detection.The serum of HAE group and control group of healthy patients summarized 19 kinds of differential metabolites.Compared with healthy people,the expression levels of Phenylacetylglycine,Tyrosine,β-Glucose,Glutamate,Lipid-7,Threonine,VLDL-2,were increased;While the expression levels of 1-Methylhistidine,Glycogen,α-Glucose,Lysine,Lipid-9,3-hydroxymethylglutaryl acid,Dimethylamine,Lipid-8,Valine,VLDL-1,VLDL-3,were decreased,(P<0.01).Conclusion: 1.Based on the combination of ~1H NMR,PLS-DA,PCA and OPLS-DA,it can provide the basis for distinguishing between HAE and healthy people,and provide a basis for further exploring the pathogenesis and diagnosis of HAE.2.HAE group and ~1H NMR group serum metabolic control group of healthy people,suggesting that the two have obvious differences in energy metabolism,amino acid metabolism,lipid metabolism,may lead to metabolic disorders in patients with HAE in the body,help to understand the mechanism of the occurrence and development of HAE. |
