Objective To investigate the effect of hydrogen sulfide on the secretion of TNF-α and IL-1β of burnt rat dermal macrophages in vitro.Methods Subjects were selected for burnt(and treated)rats with dorsal wound dermal macrophages.There are 30 healthy SD rats(male or female),5 of them are selected for normal feeding,the others are burned and fed.One of the nomal feeding rats was randomly selected as normal control group(Group 1).Twelve of the burned and fed rats were randomly divided into four groups:Burnt control group(Group 2)Na HS(exogenous H2 S donor)group 1h,6h,12h(Group3)Glibenclamide group(KATP channel blocker)1h,6h,12h(Group4)Na HS+glibenclamide group 1h,6h,12h(Group 5).The dorsum of the rats in burnt groups were made into deep II ° 5% TBSA burnt injury(wound pathology confirmed)model group.The rats in the normal control group were not treated.The dermal macrophages from the dorsal skin of the normal control group and the burnt rat wounds were cultured in vitro.During the culture process,Group 1 & 2 macrophages were added into PBS powdered,group 3were added into the Na HS solution with a concentration of 200μmol/L,group 4were added into the Gli solution with a concentration of 200μmol/L,Group 5 were added into the Na HS+Glisolution,with a concentration of 200μmol/L each.The levels of TNF-α and IL-1β in supernatant of macrophageswere identified by ELISA at +1h,+6h and +12h after adding reagent.Results At each comparable point within the whole monitoring process,the levels of TNF-α and IL-1β in the burnt control group macrophages were higher than those in the normal control group at each time point,the difference was statistically significant(P <0.05).The levels of TNF-α and IL-1β in the Na HS group were lower than those in the burnt control group,the difference was statistically significant(P <0.05).The levels of TNF-α and IL-1β in the Gli group were higher than those in the burnt control group,the difference was statistically significant(P <0.05).The levels of TNF-α and IL-1β in the Na HS group were lower than those in the Gli group,the difference was statistically significant(P <0.05).The levels of TNF-α and IL-1β in the Na HS group were lower than those in the Na HS+Gli group,the difference was statistically significant(P <0.05).The levels of TNF-α and IL-1β in the Gli group were higher than those in the Na HS+Gli group,the difference was statistically significant(P <0.05).Conclusion Exogenous H2 S can inhibit the secretion of inflammatory mediators TNF-α and IL-1β in burnt wound macrophages,which will in turn inhibit the extent of inflammatory reaction after burn.H2 S can exert its physiological regulator function in macrophages through KATP channel,and glibenclamide can prevent the H2 S from producing biological effects by controlling the KATP channel.
