Objective To investigate and study the effect of hyaluronic acid(HA)on the expression of interleukin-6(IL-6),IL-8 and tumor necrosis factor alpha(TNF-α)in human aortic vascular smooth muscle cells(HA-VSMC)induced by free fatty acids(FFA).Methods HA-VSMC cultured in vitro were treated with different concentrations of FFA(0,100,200,400 μmol / L)and FFA + HA [include high molecular weight hyaluronic acid(HMW-HA)and low molecular weight hyaluronic acid(LMW-HA),1mg / ml] for 24,48h;The effect of FFA and FFA +HA on the survival rate of HA-VSMC cells was detected by MTT method;Cell proliferation activity was detected by Ed U staining;ELISA kit for the detection of IL-6,IL-8,TNF-α expression;real-time fluorescence quantitative PCR and Western blot method was used to detect the IL-6,IL-8 and TNF-α m RNA and protein levels of expression.Results(1)FFA in 400μmol/L could induce HA-VSMC proliferation and there is time and concentration-dependent manner;(2)HMW-HA and LMW-HA have inhibitory effect on FFA induced HA-VSMC proliferation,and the inhibition of HMW-HA is stronger than LMW-HA;(3)FFA alone and in combination with the high and low molecular weight HA treatment cells 6,12,24,48 h after the cell culture supernatant IL-6,IL-8 and TNF-α with the increase of the concentration and time of culture,HA can inhibit the above effect;(4)fluorescence quantitativePCR and Western blot results showed that FFA + HMW-HA group and FFA+LMW-HA group,IL-6,IL-8 and TNF-α m RNA and protein expression level was lower than that in the FFA group,the group FFA + HMW-HA is lower than group FFA+LMW-HA,the difference had statistical significance(P<0.01).Conclusions(1)Free fatty acids in 400μmol / L can induce HA-VSMC proliferation and concentration-dependent.(2)HA could inhibit the proliferation of HA-VSMC induced by FFA,and the inhibition of HMW-HA was stronger than that of LMW-HA.(3)FFA significantly promoted the secretion of IL-6,IL-8 and TNF-α in HA-VSMC,and had significant time and concentration tolerance.(4)FFA induced the expression of IL-6,IL-8 and TNF-α m RNA and protein in HA-VSMC cells.(5)HA inhibited the expression of IL-6,IL-8 and TNF-α in HA-VSMC cells induced by FFA,down-regulated IL-6,IL-8 and TNF-α m RNA and protein expression,and HMW-HA inhibitory effect is stronger than LMW-HA.
