| Objective: To investigate the effect of micro RNA-155(mi R-155)on sorafenib resistance in hepatocellular carcinoma(HCC).Methods:Lentivirus mediated mi R-155 inhibition was transfected into SMMC-7721 cells,while lentivirus mediated mi R-155 overexpression was transfected into Hep G2 cells.The level of mi R-155 was evaluated by q PCR.Cell viability and apoptosis were analyzed by Cell Counting kit 8(CCK 8)assay and flow cytometry,respectively.The protein expression of activated caspase-3 was measured by Western blot.Results:Compared to control group,the expression of mi R-155 was significantly downregulated in mi R-155 inhibition leutivirus infected SMMC-7721 cells(P<0.01),sorafenib treatment markedly suppressed cell viability(P < 0.05)and increased cell apoptosis(P < 0.01),as well as enhanced the expression of activated caspase-3(P<0.01).However,Hep G2 cells were infected by mi R-155 overexpression leutivirus which deserved completely opposite results.Conclusion:MiR-155 may participate in sorafenib resistance in hepatocellular carcinoma and provide promising molecular target for the treatment of HCC. |
