Objective:To investigate the expression of tumor-associated macrophages(TAMs)and matrix metalloproteinase-2(MMP-2)in ameloblastomas(ABs),analysis the correlation between them and the relation with microvessel density(MVD).To preliminary discuss their mechanism in ABs’ invasive behavior.Methods:Samples of 43 paraffin-embedded ABs were collected,in addition 10 normal oral mucosa were collected as control.The expression of TAMs(marked by CD68)、MMP-2and MVD(marked by CD34)was examined by universal two-step IHC technique.Results:1.The counts of TAMs in ABs were 27.48±17.64,mainly expressed within the stroma.The MMP-2 was expressed in all 43 ABs with different intensity,mainly expressed within the islands of tumor.The counts of MVD in ABs were 21.84±8.20.2.Expression of TAMs、MMP-2 and MVD were significantly higher in ABs compared with normal oral mucosa tissue(P<0.01).3.Spearman rank correlation analysis revealed a positive correlation between MMP-2and TAMs in ABs(rs=0.350,P<0.05).The counts of MVD were positive correlated with TAMs counts and MMP-2 intensity(r=0.454,P<0.01;rs=0.459,P<0.01).Conclusion:1.There was an obvious expression of both TAMs and MMP-2 in ABs,they may participate in ABs’ development;2.There was a correlation between TAMs and MMP-2 in ABs,which may have a synergistic effect;3.The counts of MVD in ABs were higher compared with normal mucosa tissue,generation of microvessel participated in ABs’ development.TAMs and MMP-2 may promote ABs’ invasive behavior via angiogenesis inducting.
