[Objective]:To investigate the AKT/HIF/VEGF abnormal activation and p-NMDAR2 B expressionin the epileptogenic zone of the focal cortical dysplasias(FCD)relative intractable epilepsy.Then to discuss their significance and relativity analysis.[Methods]:1、Choose the 50 cases as the experimental group that was confirmed the FCD related intractable epilepsy during the 2010.01-2015 in the first hospital affiliated to Fujian Medical University,Of which includes 30 cases of FCDI,20 cases of FCDII and 8 cases of normal brain tissue from which surgical removal of intracranial decompression and brain tissue debridement.We detected the expression and protein level of P-AKT1/2/3(Thr308),HIF-1 a,VEGF,P-NMDAR2B(Tyr1336)in the brain tissue through the EIA and Western blot.[Results]:1 、 immunohistochemistry : ①P-AKT1/2/3(Thr308),HIF-1α,VEGF and P-NMDAR2B(Tyr1336)show the weak positive staining or negative expression in small amount of pyramidal neurons of the normal tissue.In the brain lesion of FCDI and FCDII,the result shows moderate positive or intense positive expression at certain degrees with a statistically significant difference(P<0.05).② P-AKT1/2/3(Thr308),P-NMDAR2B(Tyr1336)in brain lesion of FCDII is significant stronger than the FCDI with a statistically significant difference(P<0.05).Whereas there is no statistically significant difference of the HIF-1α,VEGF in the FCDI amd FCDII brain lesion.2、Western blotting:①The protein level of the P-AKT1/2/3(Thr308),HIF-1α,VEGF in the FCDI,FCDII brain lesion are significant higher than the normal brain tissue,with a statistically significant difference.②The protein level of P-AKT1/2/3/(thr308),P-NMDAR2B(Tyr1336)in FCDII brain tissue is much higher than FCDI lesion.with the significant difference.whereas there is no significant difference of the HIF-1α,VEGF between the FCDI and FCDII.[conclusion]:1、AKT/HIF/VEGF signal path was activated in some extent in the different kinds of epileptogenic zone of the focal cortical dysplasias relative intractable epilepsy.This may be the cause of the histopathologic change and the pathogenesis of spontaneous recurrent seizure.2、There is abnormal expression in the epileptogenic zone of the focal cortical dysplasias relative intractable epilepsy.It has the facilitatory effect to the neuronal excitability that leading to the epilepsy.the expression of the P-NMDAR2 B issynchronized with the severity of the pathological damage in the FCD zone.The differential expression may be the susceptibility of epilepsy.3 、 AKT/HIF/VEGF signal path may control the NMDA receptor activity and epileptogenic pathogenesis.
