Objective: The purpose of this study is to compare the cytotoxic and cell adhesion effects of a self-adhesive flowable resin composite Vertise Flow with the nano-composite flowable resin composite Filtek TMZ350 XT in vitro experiment.Methods:(1)The human pulp cells and gingival fibroblasts were collected and cultured from healthy human dental pulp and gingiva by explant combined with trypsin-digested technique.Specimens from two flowable resin composites(Vertise Flow and Filtek TMZ350XT)were eluted with culture medium,Diluted solutions of extracts at 50%、25% and 12.5% concentrations were prepared.(2)After 24 h of incubation,cytotoxicity of elutes with four concentrations were evaluated by CCK-8 assay;(3)After 72 h of incubation,apoptosis was analyzed by flow cytometry with using the Annexin-V-FITC and PI;(4)Growth of human pulp cells and gingival fibroblasts cultured in extracts with the material were observed by SEM.Result:(1)Cytotoxical grades of Vertise Flow at original concentration was grade 2,the others of Vertise Flow were grade 1;Cytotoxical grades of Filtek TMZ350 XT at every concentration were grade 1;(2)Vertise Flow and Filtek TMZ350 XT induced human dental pulp cells and gingival fibroblasts apoptosis,early apoptosis was the main form of the cell death;Compared with negative control group,statistically differences in the rate of early apoptosis were found in Vertise Flow group for these two kinds of cells,the rate of late apoptosis was only found for the human gingival fibroblasts;statistically differences in the rate of early apoptosis were found in Filtek TMZ350 XT group only for the human gingival fibroblasts,no statistically differences were found in the rate of late apoptosis.(3)A small number of human dental pulp cells and gingival fibroblasts cultured in the medium with the mode of Vertise Flow could attach to the glass substrate,morphologically,part of these cells were small-size and round-shaped;the cells cultured in the Filtek TMZ350 XT group presented numerous cells adhering to the glass and kept spindle-shaped.Conclusions: The self-adhesive flowable resin composite Vertise Flow and nano-composite flowable resin composite Filtek TMZ350 XT have somewhat cytotoxicity;Vertise Flow induced more cytotoxicity than Filtek TMZ350 XT.
