Objective: Explore the association of tumor necrosis factor alpha(TNF alpha)、 interleukin 1(IL-1)and interleukin 8(IL-8)serum level changes and the progression of pancreatic encephalopathy after using the p38 MAPK inhibitor SB203580 in rats of severe acute pancreatitis complicated with cerebral damage.analyze its relevance to the progression of pancreatic encephalopathy,for clinical diagnosis and treatment of pancreatic encephalopathy to provide experimental basis.Methods: Select 70 SD rats were randomly divided into control group(n = 10)、severe acute pancreatitis complicated with cerebral damage group(SAP group,n = 30)and SB203580 treatment group(SB group,n = 30),the rats of SAP group and SB group were injected caerulein into intraperitoneal 6 times,each time 1 hour interval,the last time lipopolysaccharide injection at the same time,induced rat severe acute pancreatitis complicated with cerebral damage models.And the rats of control group were injected the same dose normal saline into intraperitoneal.The rats of SB group before building models 0.5h were injected p38 MAPK inhibitor SB203580 into caudal vein,rats of each group in building models 3,6,12 hours were executed only 10 rats in every time span(the rats of control group were executed in building models 12 hours).Take SAP group 、SB group and control group rats brain tissue and pancreatic tissue.Observation of brain cells and nerve fibers,pancreatic tissue pathological changes.TNF-α content,IL-1 content and IL-8 content were measured in the serum in rats.Results: With the extension of time,SAP group and SB group comparison to control group changes significantly in the aspects of TNF-α content、IL-8 content and edema of nerve cells(P,all<0.05),SAP group comparison to control group changes significantly in the aspect of IL-1 content(P,all<0.05).The building after 3,6,12 hours using the two comparison of LSD-t test.Prove the illness development,each factor show different changes.Conclusion: The method of injection caerulein and lipopolysaccharide into intraperitoneal can establish a stable model of PE.With the extension of time,The content of serum amylase、TNF-α、IL-1 and IL-8 in the serum showed a trend of increase.With the extension of time,The rat brain tissue and pancreatic tissue pathological changes showed a trend of deterioration.Application SB203580 can reduce the content of serum amylase、TNF-α,IL-1 and IL-8 in the serum,and alleviate the pathological changes of pancreatic tissue and brain tissue.Its mechanism is probably inhibit p38 MAPK signal transduction pathways.
