| Objective Optimize the extraction process of polysaccharide and flavonoids from pemphis acidula,and study its anti-inflammatory and antioxidant activity in vitro.Methods(1)PAP extracted by hot water extraction and ethanol extraction,separation and purification by TCA method to removing protein,then got PAP;the scavenging rate and reducing ability of DPPH free radical,hydroxyl radical and superoxide anion free radical were determined,and Vc was used as the positive control.The extraction of PATF: the 80% ethanol solution was used to extract PATF from pemphis acidula by ultrasonic assisted extraction.the activity of PAP and PATF: through the establishment of the mouse model of enteritis,the blood and the antioxidant function of the colon were measured,and the anti-inflammatory effects of PAP and flavonoids were investigated.Results(1)Through the method of hot water and ethanol extraction to get PAP and PATF,use TCA to remove the protein.The extraction rate was 2.31%.The protein removal rate was 79.3%.The optimum extraction process of PAP is: extraction time is 120 min,the material water ratio is 25?1,the ultrasonic power is 150 W,the extraction temperature is 80 °C.(2)Through the method of ultrasonic assisted extraction to get PATF,The optimum extraction process of PATF is: ethanol concentration was 80 %,the extraction temperature was 40 °C,the extraction time was 90 min,the ratio of material to water was about 20?1.(3)The order of antioxidant activity is PAEE>PAEEA>PAEW>PAEA.(4)PAP and PATF have in vivo anti-inflammatory activity,and the activity of PATF from pemphis acidula good than PAP,high doses of PATF has certain physiological toxicity.Conclusion PAP and PATF were obtained by hot water and alcohol precipitation method,and ultrasonic assisted method,and optimized its extraction process.PAP and PATF all have anti-inflammatory activity and good antioxidant activity,has a certain reference value for the development of new drugs and functional products about pemphis acidula. |
