ObjectiveInvestigate the expression and effects of IL-37 in primary Sj?gren syndrome.MethodsCollecting the blood samples from 30 primary Sj?gren syndrome patients and 15 healthy control,ELISA detect serum IL-37 protein level,QT-PCR detect IL-37 mRNA expression in PBMCs.Collecting the clinic information,analyze the correlations between serum IL-37 protein levels with disease activity,laboratory values and inflammatory cytokines key to primary Sj?gren syndrome.Identify recombinant human IL-37 fusion protein,QT-PCR detect the changes of IL-4、IL-12、IFN-γ mRNA expression in PBMCs after recombinant human IL-37 protein stimulation.ResultsThe mRNA and protein level expression of IL-37 in primary Sj?gren syndrome patients were significantly higher than healthy controls.Serum IL-37 protein expression were correlated with ESSDAI.The IL-37 m RNA expression were positive with levels of IL-4 、 IL-12 、IFN-γmRNA.The mRNA levels of IL-4、IL-12、IFN-γ in primary Sj?gren syndrome patients were increased compared to healthy controls,however,after recombinant IL-37 stimulation,the expression of IL-4、IL-12、IFN-γmRNA in primary Sj?gren syndrome patients were obviously inhibited.Conclusions1.The high expression of IL-37 in primary Sj?gren syndrome patients suggesting IL-37 may participate in the onset of primary Sj?gren syndrome.2.IL-37 is closely related with primary Sj?gren syndrome disease activity and inflammatory cytokines,suggesting inflammatory reaction are benefit for IL-37 production.3.IL-37 inhibits the inflammatory cytokines expressions of primary Sj?gren syndrome patients,suggesting IL-37 can alleviate inflammatory reaction on the diease.
