[Objective]The aim of this study was to analysis differences in respiratory tract flora between children with bronchial asthma and normal children[method]6-14 years old children were divided into three groups, and each group has 20 people, group A:acute attack of bronchial asthma group;group B:The children above after a week’s symptomatic treatment, the symptoms were relieved as symptom remission group;group C:the control group was normal children.Throat swabs were taken from all children. After bacterial DNAs were extracted from the samples and examined by PCR of 16S rDNA V3 Region, bacterial community profilings were obtained by the use of denaturing gradient gel electrophoresis(DGGE).Quantity One and Biodap softwares were used to analyze diversity and similarity differences of respiratory tract microbial structures among three groups.[result]1.The Stripe number(S’),Shannon-Weaver Diversity Index(H’),Pielou Index(E)and Simpson index(D)of respiratory tract microflora of each phase in group A were significantly higher than those of group C(P<0.05).2.The Stripe number(S’)and Shannon-Weaver Diversity Index(H’)of respiratory tract microflora in group A were significantly higher than those in group B,and the Pielou Index(E)and Simpson index(D)were not statistically significant compared to those in group B.3.The differences of the Stripe number(S’),Shannon-Weaver Diversity Index(H’),Pielou Index(E)and Simpson index(D)between group B and group C were not significant(P>0.05).[Conclusion]:1,In the acute exacerbation group, the total number of bacteria was more abundant, the species was more, and the dominant flora was more prominent.2,Compared to the symptom remission group, the Stripe number and the Shannon-Weaver Diversity Index were higher in the acute attack of bronchial asthma group, but the Pielou Index(E)and the Simpson indexhad no differences in the two groups, and it may be associated with the use of anti-infective drugs and hormones, which caused the micro-ecological imbalance of respiratory tract.3,There was no statistically significant difference in the diversity of respiratory tract flora between normal children and children in symptom remission group, may be it was related to the partial use of anti-infective drugs and/or hormones,as well as microbial flora has the function of self-regulating balance.
